This multicenter phase II trial evaluated the therapeutic activity and safety profile of pivaloyloxymethyl butyrate (Pivanex, AN-9) as a single agent in refractory non-small cell lung cancer (NSCLC). Pivanex (2.34 g/m2 per day) was administered as a 6-h continuous intravenous infusion, daily for 3 days, and repeated every 21 days until disease progression. Forty-seven patients were treated. More than 90% of patients had received both a platinum compound and a taxane and 32% had received three or more prior chemotherapy regimens. The most common toxicities were transient grade 1-2 fatigue (34%), nausea (17%), and dysgeusia (11%). Three patients had partial responses (6.4 and 95%; CI 1.4-18.7%) and 14 patients had stable disease for > or =12 weeks (30%). Median survival for all patients was 6.2 months with 1-year survival of 26%. For patients who received fewer than three prior chemotherapy regimens, median survival was 7.8 months and 1-year survival was 31%. Pivanex is well tolerated and appears to be active as a single agent in patients with advanced NSCLC refractory to previous chemotherapy. Based on its therapeutic activity and favorable safety profile, further studies of Pivanex in NSCLC, particularly in combination with current chemotherapeutic agents, are warranted.

Chukrasia velutina is a local medicinal plant commonly known as chikrassy in Bangladesh, India, China, and other South Asian countries. The leaves, bark, and seeds are vastly used as herbal medicine for fever and diarrhea, and its leaves essential oils are used for antimicrobial purposes. In this study, we discuss the neuropsychiatric properties of C. velutina leaves through several animal models, quant. and qual. phytochem. anal., and computational approaches. Neuropsychiatric effects were performed in rodents on the methanolic extract of C. velutina leaves (MECVL). Antidepressant, anxiolytic, and sedative effects experimented through these rodent models were used such as the force swimming test (FST), tail suspension test (TST), hole board test (HBT), elevated plus maze test (EPMT), light/dark box test (LDBT), open field test (OFT), and hole cross test (HCT). In these rodent models, 200 and 400 mg/kg doses were used which exhibited a significant result in the force swimming and tail suspension test (p < 0.001) for the antidepressant effect. In the anxiolytic study, the results were significant in the hole board, elevated plus maze, and light/dark box test (p < 0.001) for doses of 200 and 400 mg/kg. The result was also significant in the open field and hole cross test (p < 0.001) for sedative action in the sake of similar doses. Moreover, qual. and quant. studies were also performed through phytochem. screening and GC-MS anal., and fifty-seven phytochem. compounds were found. These compounds were analyzed for pharmacokinetics properties using the SwissADME tool and from them, thirty-five compounds were considered for the mol. docking anal. These phytoconstituents were docking against the human serotonin receptor, potassium channel receptor, and crystal structure of human beta-receptor, where eight of the compounds showed a good binding affinity towards the resp. receptors considered to the reference standard drugs. After all of these analyses, it can be said that the secondary metabolite of C. velutina leaves (MECVL) could be a good source for inhibiting the neuropsychiatric disorders which were found on animal models as well as in computational studies.

Novel syntheses of bifunctional liquid-crystalline acrylate and methacrylate monomers are described. Thermal and microstructural characterization of the monomers using small-angle X-ray scattering, differential scanning calorimetry, and optical microscopy revealed the presence of smectic A, smectic C, and smectic E phases. Photopolymerization of the bifunctional monomers conducted at different temperatures preserved the monomeric liquid-crystalline structure in the crosslinked polymers. The densely crosslinked polymers showed thermal reversibility. Mixtures of these monomers and chiral monofunctional liquid-crystalline monomers exhibited chiral smectic C mesomorphism that was made permanent by photopolymerization and subsequent crosslinking.

We previously reported that lysine-demethylase 2A (KDM2A), a Jumonji-C histone demethylase, is activated by gallic acid to reduce H3K36me2 levels in the rRNA gene promoter and consequently inhibit rRNA transcription and cell proliferation in the breast cancer cell line MCF-7. Gallic acid activates AMP-activated protein kinase (AMPK) and increases reactive oxygen species (ROS) production to activate KDM2A. Esters of gallic acid, Pr gallate (PG) and epigallocatechin gallate (EGCG), and other chems., reduce cancer cell proliferation. However, whether these compounds activate KDM2A has yet to be tested. In this study, we found that PG and EGCG decreased rRNA transcription and cell proliferation through KDM2A in MCF-7 cells. The activation of both AMPK and ROS production by PG or EGCG was required to activate KDM2A. Of note, while the elevation of ROS production by PG or EGCG was limited in time, it was sufficient to activate KDM2A. Importantly, the inhibition of rRNA transcription and cell proliferation by gallic acid, PG, or EGCG was specifically observed in MCF-7 cells, whereas it was not observed in non-tumorigenic MCF10A cells. Altogether, these results suggest that the derivatization of gallic acid may be used to obtain new compounds with anti-cancer activity.

This is the first account of a triply unsaturated pheromone component in a tortricid moth. The monoenic acetate (E)-7-dodecenyl acetate and the trienic acetate (7Z,9E,11)-dodecatrienyl acetate significantly enhanced responses of males to the main pheromone compound, (7E,9Z)-7,9-dodecadienyl acetate, in the wind tunnel. The identification of sex pheromone synergists in L. botrana may be of practical importance for the development of integrated pest management systems.

Archips atrolucens, Adoxophyes privatana, and Homona sp. are serious defoliators of citrus trees in the Mekong Delta of Vietnam. To establish a sustainable pest-management program for the 3 species, their female-produced sex pheromones were investigated by GC-EAD and GC-MS analyses, and the following multi-component pheromones were identified: (Z)-11-tetradecenyl acetate (Z11-14:OAc), (E)-11-tetradecenyl acetate (E11-14:OAc), and tetradecyl acetate (14:OAc) in a ratio of 64:32:4 for A. atrolucens; Z11-14:OAc and (Z)-9-tetradecenyl acetate (Z9-14:OAc) in a ratio of 92:8 for A. privatana; and Z11-14:OAc and (Z)-9-dodecenyl acetate (Z9-12:OAc) in a ratio of 96:4 for Homona sp. Each lure baited with synthetic components as a mimic of the natural pheromone attracted males of the target species specifically, indicating that each monounsaturated minor component plays a significant role for mating communication and reproductive isolation of the three species inhabiting the same citrus orchards. In an extract of the pheromone glands of A. atrolucens females, the content of 14:OAc was very low, but a synergistic effect was observed clearly when the saturated compound was mixed at the same level as the E11-14:OAc. The synthetic lures will provide useful tools for monitoring flights of adults of the 3 species.

An electromagnetic energy source in the radio-frequency range delivers power to a stent circuit via resonant inductive coupling, allowing a thermally triggered release of gel via Joule heating. A gold-electroplated, medical-grade stainless steel stent, serving as the base of the prototype device, melts a coating made from an emulsion composed mainly of dodecanoic acid. These coated devices produce wirelessly controllable releases of a gel into thermally regulated, stirred water that is near body temperature The gel is made from salt, water, and gelatine from porcine skin and used to simulate drug release in this study. Thus, this system serves as a proof of concept to show the viability of controlling local drug delivery using this prototype device. Dodecanoic acid, a fatty acid, has a phase transition from solid to liquid near 43°C and has relatively good biocompatibility. The average melting temperature of two different emulsions was 40.8 ± 0.7°C, a suitable value for the targeted application. Demonstration of controllable releases used electromagnetic pulses of approx. 180 s in duration, illustrating reproducibility of a controllable release phase while remaining relatively inert in the absence of stimuli. Releases were observable through measuring the conductivity of the water, the water temperature, and the stent temperature This electrothermally active stent device enables wirelessly controlled local delivery with controlled dosage and timing, a concept with a wide range of potential applications. Some relevant examples include inhibiting restenosis or cancer treatment via targeted chemotherapy.
