A simple, one-pot synthesis of a surface-tethered MADIX agent was established via the reaction of 1,1′-thiocarbonyl diimidazole (TCDI) with a thiol, followed by reaction with the hydroxyl groups of a Wang resin. The supported RAFT agent was used to control the polymerization of vinyl acetate in toluene. The amount of surface-bound polymer was found to greatly increase with increasing content of solid MADIX agent, reaching values of almost 80%. The grafted PVAc chains were cleaved from the Wang resin support via radical reaction with tert-amyl peroxy acetate (TAPA), and the mol. weight distributions exhibited the typical shape of polymers formed via a well-controlled living radical polymerization (LRP), with the free polymers showing a slightly higher mol. weight than the living fraction. The Mn values of both living polymer fraction (surface-bound polymer) and dead polymer (free polymer) increased steadily with monomer conversion-as expected for a well controlled MADIX polymerization-and were in good agreement with the theor. mol. weights PDI values of both polymer fractions scatter around 1.5, with a slight tendency of narrower mol. weight distributions of the living polymer, which shows a min. value of 1.43. Addition of free MADIX agent to the solution phase led to a decrease in PDI values of both free and bound polymer, and the exptl. molar masses were in good agreement with theor. predictions.

The desfluoroquinolone-based hybrids with involvement of C-7 aminopyrimidine functional group were designed and synthesized. The biol. results showed majority of these hybrids still demonstrated potent anti-MRSA activity with MIC values between 0.38 and 1.5μg/mL, despite the lack of the typical C-6 fluorine atom. Particularly, the most active 1-cyclopropyl-7-((4-(3,4-dimethylphenoxy)pyrimidin-2-yl) amino)-4-oxo-1,4-dihydroquinoline-3-carboxylic acid exhibited activities at submicromolar concentrations against a panel of MRSA strains including vancomycin-intermediate strains, levofloxacin-resistant isolates, and linezolid-resistant isolates, etc. As expected, it also displayed highly selective toxicity toward bacterial cells and low hERG inhibition. Further resistance development study indicated MRSA is unlikely acquired resistance against 1-cyclopropyl-7-((4-(3,4-dimethylphenoxy)pyrimidin-2-yl)amino)-4-oxo-1,4-dihydroquinoline-3-carboxylic acid. The docking study revealed that two hydrogen bonds were formed between the C-7 substituent and the surrounding DNA bases, which contributed to resistance by reducing the dependence on the magnesium-water bridge interactions with topoisomerase IV. These indicated a promising strategy for developing new antibiotic quinolones to combat multidrug resistance and cardiotoxicity was resulted.

A novel strategy for the synthesis of main-chain polymers through radical ring-closing/ring-opening cascade polymerization is reported. Efficient radical cyclopolymn. was achieved through systematic optimization of the electronic properties of 1,6-diene structures. Fusing 1,6-diene with allylic sulfide or allylic sulfone motifs enabled a ring-closing/ring-opening cascade reaction that provides a strong driving force for the ring-opening polymerization of large macrocyclic monomers. The ability of 1,6-diene-fused allylic sulfone to undergo efficient SO2 extrusion generated a propagating alkyl radical capable of reversible deactivation. This strategy provides a general platform for the synthesis of polymers incorporating complex main-chain structures and degradable functionalities.

Low-mol.-weight (LMW) thiols are important small mols. that regulate or maintain redox homeostasis in physiol. and pathol. processes. Assessing the concentrations of LMW thiols in biol. systems may provide valuable information regarding physiol. processes and the early diagnosis of some diseases. Here, we developed a method to simultaneously determine the concentrations of multiple LWM thiols in single cells by chem. derivatization assisted liquid chromatog.-mass spectrometry (LC-MS). In this method, we synthesized a pair of stable isotope reagents, N-(acridin-9-yl)-2-bromoacetamide (AYBA) and N-(1,2,3,4-[2H4]-acridin-9-yl)-2-bromoacetamide ([2H4]AYBA). AYBA was used to derivatize LWM thiols in human cervical cancer (HeLa) cells, while [2H4]AYBA was used to derivatize standard LWM thiols to prepare internal standards for the LC-MS method development. The proposed AYBA derivatization greatly enhanced the detection sensitivity of LWM thiols by LC-MS, and thereby achieved the simultaneous detection of multiple LWM thiols by LC-MS in ∼1000 HeLa cells. Finally, the developed method was successfully utilized for the quant. anal. of multiple LWM thiols in a single HeLa cell and the content changes of LWM thiols in a single HeLa cell before and after oxidative stress treatment. Accordingly, six LMW thiols were detected, including cysteamine, cysteine, glutathione, homocysteine, hydrogen sulfide, and pantetheine.

BACKGROUND: Dimethyl fumarate (DMF) is an effective drug for multiple sclerosis and can improve the cognitive dysfunction caused by streptozotocin, but the effect on cognitive dysfunction caused by hypothyroidism is unclear. METHODS: After the hypothyroidism rat model induced by propylthiouracil, we gave rats 25 mg/kg DMF by gavage. The body weight during model building and administration was recorded. The levels of T4 and T3 in serum were detected by an automatic biochemical analyzer. Morris water maze test was used to detect the effect of DMF on cognitive learning ability. The effect of DMF on Nissl bodies in the brain tissue was evaluated by Nissl staining. The mRNA and protein levels of BDNF in brain tissue were detected by quantitative reverse transcription-polymerase chain reaction and Western blot. The degrees of p-AKT/AKT and p-CREB/CREB in brain tissue were detected by Western blot. RESULTS: After DMF treatment, the body weight of hypothyroid rats recovered, and the levels of T3 and T4 in the serum were ameliorated. DMF also reduced the escape latency and distance traveled, and increased the swim speed. The number of Nissl bodies and expression of BDNF, p-AKT/AKT, and p-CREB/CREB in the brain tissue were increased after DMF treatment. CONCLUSION: DMF improved the cognitive dysfunction of hypothyroid rats by increasing the level of BDNF in the brain tissue of hypothyroid rats.

INTRODUCTION AND HYPOTHESIS: Mechanical trauma and oxidative injury are involved in the pathogenesis of stress urinary incontinence (SUI), and oxidative stress (OS) is considered a potential therapeutic target. The antioxidant properties of dimethyl fumarate (DMF), a potent activator of Nrf2, have been highlighted recently. We therefore predicted that DMF might have therapeutic effects on mechanical trauma-induced SUI. METHODS: The SUI mice model was established by vaginal distension (VD). Leak point pressure (LPP), serum OS biomarkers, cell proliferation and apoptosis, collagen, elastin, matrix metalloproteinases (MMP), Nrf2, the TGF-β1/Smad3 signaling pathway, and the associated tissue growth factors in the anterior vaginal wall were measured in either wild-type or Nrf2-knockout (Nrf2/) female C57BL/6 mice. RESULTS: The results showed that DMF improved the VD-induced LPP reduction, alleviated oxidative injury, stimulated cell proliferation and inhibited apoptosis in the anterior vaginal wall tissue of mice. Moreover, DMF treatment reduced the hydrolysis of ECM proteins by MMP2 and MMP9. The above effects may be mediated by a series of tissue growth factors, including α-SMA, PAI-1, and TIMP-2, with the TGF-β1/Smad3 signaling pathway as the core regulatory mechanism. In further study, Nrf2/ mice were used to replicate the SUI model. And the difference is that DMF failed to reactivate the TGF-β1/Smad3 pathway, nor did it improve LPP. CONCLUSIONS: Dimethyl fumarate can ameliorate urethra closure dysfunction in the VD-induced SUI mice model, and the therapeutic effect of DMF is mediated by the Nrf2-dominated antioxidant system and its downstream TGF-β1/Smad3 signaling pathway.

High-performance stimuli-responsive polymers that exhibit spontaneous, sophisticated and reversible responses to a wide range of external stimuli are highly desirable for applications in diverse fields and have attracted dramatic attention in recent decades. However, it still remains a great challenge to precisely design mol. architectures which can integrate multi-stimuli responsive functions into one structure in polymeric materials. In this study, we reported the synthesis and preparation of a novel multi-responsive self-healing polyurethane with superior mech. properties by adapting a stimuli-responsive dynamic covalent chem. crosslinker and a biomimetic modular polymer design. This unique crosslinker was synthesized through the Michael addition reaction between thiols and bismaleimides, making the chem. crosslinked polyurethanes healable and recyclable at an elevated temperature as well as stimuli-responsive to heat, pH and ammonia gas. A visible color change can be clearly observed when the polymer is in the base or ammonia gas environment, giving the sample great potential for being a gas or pH sensor. Furthermore, the incorporation of self-complementary quadruple hydrogen bonding interactions between 2-ureido-4-[1H]-pyrimidione (UPy) motifs in the polyurethane backbone chains enables the formation of modular hydrogen bonding domains, endowing the material with enhanced mech. properties.
