Porey, Arka;Santra, Surojit;Guin, Joyram research published 《 Highly Enantioselective Synthesis of Functionalized Glutarimide Using Oxidative N-Heterocyclic Carbene Catalysis: A Formal Synthesis of (-)-Paroxetine》, the research content is summarized as follows. A simple yet highly effective approach toward enantioselective synthesis of trans-3,4-disubstituted glutarimides from readily available starting materials is developed using oxidative N-heterocyclic carbene catalysis. The catalytic reaction involves a formal [3 + 3] annulation between enals and substituted malonamides enabling the production of glutarimide derivatives in a single chem. operation via concomitant formation of C-C and C-N bonds. The reaction offers easy access to a broad range of functionalized glutarimides with excellent enantioselectivity and good yield. Synthetic application of the method is demonstrated via formal synthesis of (-)-paroxetine and other bioactive mols.

Plamondon, Samuel J.;Warnica, Josephine M.;Kaldre, Dainis;Gleason, James L. research published 《 Hydrazide-Catalyzed Polyene Cyclization: Asymmetric Organocatalytic Synthesis of cis-Decalins》, the research content is summarized as follows. Polyene cyclizations offer rapid entry into terpenoid ring systems. Although enantioselective cyclizations of (E)-polyenes to form trans-decalin ring systems are well precedented, highly enantioselective cyclizations of (Z)-polyenes to form the corresponding cis-decalins have not been reported. Here, we describe the first application of iminium catalysis to the initiation of polyene cyclizations. Et 1,2-diazepane-1-carboxylate catalyzes the cyclization of polyenes bearing enal initiators. Moreover, chiral bicyclic hydrazides catalyze the cyclizations of (Z)-polyene substrates to form cis-decalins with enantioselectivities of up to 97:3 er. DFT calculations suggest the catalysts promote the reaction by stabilizing pos. charge as it develops during the bicyclization.

Protein-protein interactions (PPIs) have emerged as significant targets for therapeutic development, owing to their critical nature in diverse biol. processes. An ideal PPI-based target is the protein myeloid cell leukemia 1 (MCL1), a critical prosurvival factor in cancers such as multiple myeloma where MCL1 levels directly correlate to disease progression. Current strategies for halting the antiapoptotic properties of MCL1 revolve around inhibiting its sequestration of proapoptotic factors. Existing inhibitors disrupt endogenous regulatory proteins; however, this strategy actually increases MCL1 protein levels. Here, the authors show the development of hetero-bifunctional small mols. capable of selectively targeting MCL1 using a proteolysis targeting chimera (PROTAC) methodol. leading to successful degradation The authors confirmed the involvement of the E3 ligase CUL4A-DDB1 cereblon ubiquitination pathway, making these PROTACs a first step toward a new class of antiapoptotic B-cell lymphoma 2 family protein degraders.

Known hydrophilic halide salts of the title compounds are converted to new lipophilic BArf (B(3,5-C6H3(CF3)2)4) salts. These are isolated as hydrates (Λ- or Δ-[M(en)3]n+nBArf·zH2O; z = 17-9) and characterized by NMR (acetone-d6) and microanalyses. Thermal stabilities are probed by capillary thermolyses and TGA and DSC measurements (onset of dehydration 71-151°C). In the presence of tertiary amines, they are effective catalysts for enantioselective Michael type carbon-carbon or carbon-nitrogen bond forming additions of 1,3-dicarbonyl compounds (acceptors: trans-β-nitrostyrene, di-tert-butylazodicarboxylate, 2-cyclopenten-1-one; average ee = 33%, 52%, 17%). Polyesters are important plastics, with monomers linked by ester moieties. Phosphoesters form the backbone of DNA molecules. 870-50-8, formula is C10H18N2O4, Name is Di-tert-butyl diazene-1,2-dicarboxylate.Nitrate esters, such as nitroglycerin, are known for their explosive properties. Category: esters-buliding-blocks.

Merchant, Rohan R.;Lang, Simon B.;Yu, Tingting;Zhao, Shuilian;Qi, Zhiqi;Suzuki, Takao;Bao, Jianming research published 《 A General One-Pot Protocol for Hindered N-Alkyl Azaheterocycles from Tertiary Carboxylic Acids》, the research content is summarized as follows. Herein, a general one-pot strategy that utilizes three elementary steps (decarboxylative hydrazination, Boc deprotection, and heterocycle condensation) to regioselectively prepare hindered C(sp3) substituted pyrazoles, e.g., I and triazole II was reported. The operational simplicity of this sequence and ubiquity of tertiary carboxylic acids allow rapid access to hindered N-alkyl azaheterocycles that will be useful to practitioners of medicinal chem. and agro-chem.
