Song, Xue;Zhang, Jie;Wu, Yu-Xing;Ouyang, Qin;Du, Wei;Chen, Ying-Chun research published 《 Asymmetric Formal Nucleophilic o-Cresolylation with Morita-Baylis-Hillman Carbonates of 2-Cyclohexenones via Palladium Catalysis》, the research content is summarized as follows. Here Authors report an asym. formal nucleophilic o-cresolylation reaction with the Morita-Baylis-Hillman (MBH) carbonates from 2-cyclohexanones and diverse aldehydes under palladium catalysis, by in situ generation of electron-neutral and HOMO-raised η2-Pd(0)-dienone complexes via an oxidative insertion/π-σ-isomerization/β-H elimination activation sequence. The subsequent umpolung vinylogous addition to a variety of imines is realized upon Pd(0)-mediated π-Lewis base catalysis, finally furnishing o-cresolylated products followed by another cascade of a π-σ-isomerization/β-H elimination/aromatization process. Moderate to excellent diastereo- and enantioselectivity are achieved for substantial substrate assemblies by employing a newly designed bulky chiral phosphonamidite ligand, and the resultant multifunctional products can be facilely elaborated to access diverse enantioenriched architectures. In addition, the catalytic reaction pathway is finely illuminated by control experiments

Shen, Wen;Cunningham, Laura;Evans, Paul research published 《 Asymmetric synthesis of γ-amino-functionalized vinyl sulfones: De novo preparation of cysteine protease inhibitors》, the research content is summarized as follows. The enantioselective azo-based α-amination of an aldehyde followed by a Horner-Wadsworth-Emmons-based vinyl sulfone formation is reported. The thus obtained optically active N, N’-diprotected trans-(phenylsulfonyl)vinyl hydrazine products were then converted into the corresponding N-functionalized trans-(phenylsulfonyl)vinyl amines. Specifically, reaction of 4-phenylbutanal with di-tert-Bu azodicarboxylate (DBAD) in the presence of L– or D-proline, followed by addition of di-Et [(phenylsulfonyl)methyl]phosphonate, gave either enantiomer of di- tert-Bu trans-1-[5-phenyl-1-(phenylsulfonyl)pent-1-en-3-yl]hydrazine-1,2-dicarboxylate. The enantiomeric excesses of the (+)- and (-)-enantiomers prepared in this manner were in the range 86-89%. The conversion of these γ-hydrazino vinyl sulfones into the corresponding γ-amino-substituted compounds was achieved following a Boc (Boc = tert-butoxycarbonyl) deprotection, Zn reduction, N-functionalization sequence. This three-step sequence was reasonably efficient (approx. 50%) and no erosion of enantiopurity was found to have taken place. The compounds accessed via this process include both enantiomers of tert-Bu trans-[5-phenyl-1-(phenylsulfonyl)pent-1-en-3-yl]carbamate and epimeric dipeptide mimetics including 4-methyl- N-{( S)-1-oxo-3-phenyl-1-[(( S, E)-5-phenyl-1-(phenylsulfonyl)pent-1-en-3-yl)amino]propan-2-yl}piperazine-1-carboxamide (also known as K777).

This developed methodol. gives an alternative and milder way to construct the C-N bond and can be explored for the formation of C-C bond to perform arylation and allylation reactions.

Scarpi, Dina;Bagni, Francesco;Faggi, Cristina;Carral-Menoyo, Asier;Gomez-Bengoa, Enrique;Occhiato, Ernesto G. research published 《 Gold(I)-Catalyzed Cycloisomerization/Hetero-Diels-Alder Reaction/Ring Opening Cascade to Functionalized Cyclopentadienes》, the research content is summarized as follows. Six- and seven-membered ring-fused, functionalized cyclopentadienes can be obtained in moderate to excellent yields by a cascade process entailing the Au(I)-catalyzed propargyl Claisen rearrangement/Nazarov cyclization of propargyl vinyl ethers, the hetero-Diels-Alder reaction with dialkylazodicarboxylates, and the spontaneous conversion of cycloaddition products into cyclopentadienes by a highly regioselective cleavage of a C-N bond. Depending on the treatment of the crude reaction mixtures, two types of products can be obtained: cyclopentadienes with pendant hydrazine and aldehyde moieties that intramolecularly react to form hemiaminals are obtained in 43-52% overall yields when the crude reaction mixtures are left over K2CO3 in a DCM solution Instead, by reducing in situ the aldehyde group just after addition of the heterodienophile, the regioselective C-N bond cleavage generates the corresponding cyclopentadienes bearing a hydrazine and an alc. appendage in excellent yields (66-82%) over four steps, all in one pot. Two examples from the latter class of compounds were also converted into ring-fused, functionalized cyclopentadienes, bearing a protected amino group, by the selective N-N cleavage of the hydrazine moiety.

Esters typically have a pleasant smell; those of low molecular weight are commonly used as fragrances and are found in essential oils and pheromones. 870-50-8, formula is C10H18N2O4, Name is Di-tert-butyl diazene-1,2-dicarboxylate. They perform as high-grade solvents for a broad array of plastics, plasticizers, resins, and lacquers, and are one of the largest classes of synthetic lubricants on the commercial market. Category: esters-buliding-blocks.

Ratni, H.;Baumann, K.;Bellotti, P.;Cook, X. A.;Green, L. G.;Luebbers, T.;Reutlinger, M.;Stepan, A. F.;Vifian, W. research published 《 Phenyl bioisosteres in medicinal chemistry: discovery of novel γ-secretase modulators as a potential treatment for Alzheimer′s disease》, the research content is summarized as follows. Ph rings are one of the most prevalent structural moieties in active pharmaceutical ingredients, even if they often contribute to poor physico-chem. properties. Herein, we propose the use of a bridged piperidine (BP) moiety as a Ph bioisostere, which could also be seen as a superior Ph alternative as it led to strongly improved drug like properties, in terms of solubility and lipophilicity. Addnl., this BP moiety compares favorably to the recently reported saturated Ph bioisosteres. We applied this concept to our γ-secretase modulator (GSM) project for the potential treatment of Alzheimer′s disease delivering clin. candidates.
