Avian pathogenic Escherichia coli (APEC) is a common bacterial pathogen infecting chickens, resulting in economic losses to the poultry industry worldwide. In particular, APEC O1, one of the most common serotypes of APEC, is considered problematic due to its zoonotic potential. Therefore, many attempts have been made to develop an effective vaccine against APEC O1. In fact, the lipopolysaccharide (LPS) O-antigen of APEC O1 has been shown to be a potent antigen for inducing specific protective immune responses. However, the detailed structure of the O-antigen of APEC O1 is not clear. The present study demonstrates the first synthesis of a pentasaccharide repeating unit of LPS derived from virulent E. coli O1 and its conjugate with BSA. ELISA tests using the semi-synthetic glycoconjugate and the APEC O1 immune chicken serum revealed that the pentasaccharide is a glycotope candidate of APEC O1, with great potential as an antigen for vaccine development.

Nimodipine attenuates dibutyl phthalate-induced learning and memory impairment in kun ming mice: An in vivo study based on bioinformatics analysis

Dibutyl phthalate (DBP), a typical representative of phthalate esters (PAEs), is used as a plasticizer in various industrial applications and has been reported to be responsible for neurobehavioral changes. Despite mounting evidence showing that nimodipine (Nim) palys a neuropharmacological and psychopharmacological role in neurons, the attenuating effects of Nim on learning and memory impairment induced by DBP exposure remain unknown. Based on bioinformatics analysis we found that the biological processes affected by both DBP and Nim may involve the calcium signaling pathway, the MAPK signaling pathway and the apoptosis pathway. The results of an in vivo study confirmed that DBP affects the levels of Ca2+-related proteins, up-regulates phosphorylated -ERK1/2 expression and results in hippocampal neuronal damage and apoptosis, whereas Nim as a Ca2+ antagonist, has a certain neuroprotective role to avoid these adverse effects. Our data suggest that Nim could be used to attenuate the learning and memory impairment in DBP-exposed mice, to down-regulate intracellular Ca2+ levels, subordinate the ERK1/2 pathway and attenuate apoptosis in hippocampal tissue.

The oxidation mechanism of phospholipids in Antarctic krill oil promoted by metal ions

Antarctic krill oil (AKO) is an emerging dietary supplement containing metal ions that influence oil oxidation. Thus, this study focuses on the effect and mechanism of metal ions on phospholipid oxidation in AKO. Firstly, AKO containing Mg2+, Mn2+, Cu2+, Fe2+ and Fe3+ (200 mu mol/kg) were prepared separately and incubated at 60 degrees C for 6 days. Peroxide value (POV) and malondialdehyde (MDA) content showed that Fe3+ exhibited the most effective prooxidative activity, with the prooxidative activity of Fe-2(SO4)(3) (water-soluble) being stronger than that of ferric oleate (FeOl, fat-soluble). In addition, phosphatidylethanolamine (PE) oxidation degree (more than 90%) was considerably greater than phosphatidylcholine (PC) oxidation degree (about 15%) in AKO. Differences in the structure of head group hindered chelation of PC with metal ions than PE due to electrostatic repulsion and steric hindrance. Therefore, PC significantly inhibited, while PE promoted, the oxidation of polyunsaturated triacylglycerol (TAG), like fish oil (p < 0.01). Modular Synthesis of Heparan Sulfate Oligosaccharides Having N-Acetyl and N-Sulfate Moieties

Heparan sulfates are structurally diverse sulfated polysaccharides that reside at the surface of all animal cells where they can interact with a multitude of proteins, thereby modulating a wide range of physiological and disease processes. We describe here a modular synthetic methodology that can provide libraries of heparan sulfate oligosaccharides that have glucosamine residues modified by different patterns of N-acetyl and N-sulfate moieties. It is based on the use of glycosyl donors that are modified at C2 by an azido- or trifluoromethylphenyl-methanimine moiety, which allowed the selective installation of alpha-glycosides. The amino protecting groups can be selectively unmasked by a reduction or acid treatment, allowing the installation of N-acetyl and N-sulfate moieties, respectively. In combination with the orthogonal hydroxyl protecting groups levulinic (Lev) ester, thexyldimethylsilyl (TDS) ether, allyloxycarbonate (Alloc), and 9-fluorenylmethyl carbonate (Fmoc), different patterns of O-sulfation can be installed. The methodology was applied to prepare four hexasaccharides that differ in the pattern of N- and O-sulfation. Poly(vinyl alcohol)-borax complex-based spray coating for the decontamination of radioactive Cs from wide-area surfaces

A spray coating based on an adsorbent/polyvinyl alcohol (PVA)-borax complex was developed to remove radioactive cesium from surfaces by adsorption for wide-area surface decontamination. Two aqueous solutions of borax and PVA containing adsorbents, namely, Prussian blue, bentonite, and sulfur-zeolite, were simultaneously sprayed on surfaces, which subsequently self-generated a gel-like coating via a borate-diol ester bond between borax and PVA. The spray coating adhered adequately to vertical surfaces for at least 1 h due to its good viscoelasticity and displayed Cs-137 removal efficiency (56.966%) twice that of a commercial strippable coating (DeconGel, 27.248%) on porous cement surfaces because the sulfur-zeolite in the coating has an excellent Cs distribution coefficient (4.768 x 10(6) mL/g). In addition, our coating could be easily removed from the surface after use by simply rinsing with water, and it left no residue, even on a porous surface such as cement, due to the reversible PVA-borax complexation. In contrast, commercial strippable coating and chemical gels leave a residue. Moreover, the used adsorbent that captured Cs-137 could be easily separated using simple filtration for a direct solidification procedure before final disposal/storage, whereas the commercial strippable coating and chemical gels require expensive treatment, such as incineration, to remove organic components. In a demonstrative test using a commercial spray-coater, the spray coating exhibited a fast spray rate (1.25 m(2)/min), high stability (> 1 h), and easy removal by simply rinsing with tap water.

A phase change material embedded composite consisting of kapok and hollow PET fibers for dynamic thermal comfort regulation

Kapok fiber, an agricultural product, is a typical cellulosic fiber with the features of thin cell wall and large lumen. Herein, a microtubule-based composite with high porosity of 98.03 % was prepared using kapok fibers and hollow PET. The kapok fiber-based composite successfully encapsulated a phase change material (PCM), that is a fatty acid ester of myristic acid and tetradecyl alcohol (MA-TD), in the microtubules by vacuum impregnation. The SEM showed that the PCM was encapsulated in the lumens of the fibers with a calculated percentage of 40.5 %. This condition provided the composite a basic thermal insulation from air pockets in the composite and a dynamic thermal insulation from the PCM. The kapok/PET/MA-TD could absorb 76.8 J/g latent heat in the comfort temperature range of 24 degrees C – 42.3 degrees C or release 77.4 J/g latent heat in the comfort temperature range of 29.7 degrees C – 10.5 degrees C. It withstood repeated laundry and exhibited robust leakage resistance to continuous heating/cooling under pressure. In addition, the composite was successfully used to dynamically regulate the temperature with obvious thermal regulating capacity in the simulated cooling/hot environments. Such material will be a robust candidate for dynamic heat regulating and thermal insulating applications.

Reduction Chemistry of Natural Pyrethrins and Preliminary Insecticidal Activity of Reduced Pyrethrins

The natural extract pyrethrum is an insecticidal oil derived from Tanacetum cinerariifolium that is commonly used in domestic and agricultural pesticides. The major constituents of the extract are the Pyrethrins, six esters that provide pyrethrum with its insecticidal properties. These Pyrethrins readily degrade through several environmental means and as such, there can be significant Pyrethrin losses during processing and long-term storage of pyrethrum-based insecticides. This work attempts to alleviate the effect of these degradative processes through the pursuit of stabilised Pyrethrins by chemically removing oxidatively sensitive functionality. Several reduced Pyrethrin analogues were produced and a method to convert the more sensitive Pyrethrins present in the pyrethrum concentrate into their respective more stable jasmolin counterparts, as a mixture with the over-reduced tetrahydropyrethrins, was developed. All other reduction processes abolished insecticidal activity against Lucilia cuprina larvae, whereas some isomerised analogues showed comparable potency with the individual natural pyrethrin esters. This work has revealed new insights into the structure-activity relationships in this unique class of insecticide.
