The occurrence and distribution of 100 priority and emerging micropollutants were investigated in sediments within the first 25 km of the Capbreton submarine area. The most predominant compounds were polycyclic aromatic hydrocarbons (PAHs), trace metals and metalloid (TMs) (e.g. mercury, lead and arsenic), synthetical musks (e.g. musk ketone, galaxolide), UV filters (e.g. octocrylene and 2-ethylhexyl 4-methoxycinnamate, EHMC) as well as some pharmaceuticals (e.g. azithromycin, acetaminophen). Enormous production of cosmetic products and its indiscriminate use tends to discharge into the aquatic environment and might threaten non-target organisms inhabiting aquatic ecosystems. In the present study, developmental toxicity of 4-methylbenzylidene camphor (4-MBC), a widely used organic UV filter in personal care products has been evaluated using zebrafish embryo-larval stages. Waterborne exposure induced developmental toxicity and deduced 2.71 mg/L as 96 h LC50 whereas embryos exposed to sub-lethal concentrations (50 and 500μg/L) caused a significant delay in hatching rate, heart rate, reduced larval length, and restricted hatchlings motility besides the axial curvature. Chronic exposure to 10 dpf resulted in significant decrease in SOD activity at 500μg/L with no changes in CAT level besides a significant increase in GST enzyme at 5μg/L concentration in 5 dpf sampled larvae. However, all the three enzymes were significantly elevated in 10 dpf larvae indicating differential oxidative stress during the stages of development. Similar trend is noticed for acetylcholine esterase enzyme activity. A concentration dependent increase in malondialdehyde content was noted in larvae sampled at 5 and 10 dpf. In addition, multixenobiotic resistance (MXR) activity inhibition, and elevated oxidative tissue damage were noticed at 5 dpf with no significant changes in 10 dpf larvae. Furthermore, immunoblot anal. confirms 4-MBC induced apoptosis in zebrafish larvae with promoted cleaved Caspase-3, Bax and inhibited Bcl-2 proteins expression. Subsequently, docking studies revealed the binding potential of 4-MBC to zebrafish Abcb4 and CYP450 8A1 proteins with the binding energy of -8.1 and -8.5 kcal/mol representing target proteins interaction and toxicity potentiation. Our results showed that 4-MBC exposure triggers oxidative stress at sub-lethal concentrations leading to apoptosis, deformities and locomotion perturbations in developing zebrafish. This is first of its kind in systematically demonstrating developmental toxicity of 4-MBC and the information shall be used for aquatic toxicity risk assessment. It was notable that the lab-made coating exhibited extraordinary stability towards hot water steam, probably because the hydrophobic groups capped on the MOF prevented water mols. from entering and deconstructing its lattice. Even after being treated with water steam under 100°C for 21 h, the extraction performance of the coating remained unchanged, and the crystal structure of the MOF maintained. Furthermore, a negligible matrix effect was observed even in the samples containing humic acid. Under the optimal extraction and thermal desorption conditions, a method for determining UV filters in aqueous samples was established, which possessed low detection limits (0.6-2.1 ng L-1), wide linear ranges (10-50000 ng L-1), good inter-fiber reproducibility (2.3-6.0%, n = 6), and satisfying intra-fiber repeatability (1.8-5.8%, n = 3). The method was successfully applied in quantifying UV filters in environmental water samples. In addition, the lab-made NH2-UiO-66(Zr)-shp-coated fiber was also suitable for the anal. of polycyclic aromatic hydrocarbons (PAHs). Here, we evaluate 2-Amino-6-(trifluoromethoxy) benzothiazole (riluzole) as an early intervention strategy for Alzheimers disease (AD), aimed at restoring glutamate neurotransmission prior to substantial Beta amyloid (Aβ) plaque accumulation and cognitive decline. Male AβPP/PS1 mice, a model of progressive cerebral amyloidosis, were treated with riluzole from 2-6 mo of age. Morris water maze, in vivo electrochem., and immunofluorescence were performed to assess cognition, glutamatergic neurotransmission, and pathol., resp., at 12 mo. Four months of prodromal riluzole treatment in AβPP/PS1 mice resulted in long-lasting procognitive effects and attenuated glutamatergic tone that was observed six months after discontinuing riluzole treatment. Riluzole-treated AβPP/PS1 mice had significant improvement in long-term memory compared to vehicle-treated AβPP/PS1 mice that was similar to normal aging C57BL/6J control mice. Furthermore, basal glutamate concentration and evoked-glutamate release levels, which were elevated in vehicle-treated AβPP/PS1 mice, were restored to levels observed in age-matched C57BL/6J mice in AβPP/PS1 mice receiving prodromal riluzole treatment. Aβ plaque accumulation was not altered with riluzole treatment. This study supports that interventions targeting the glutamatergic system during the early stages of AD progression have long-term effects on disease outcome, and importantly may prevent cognitive decline. Our observations provide preclin. support for targeting glutamate neurotransmission in patients at risk for developing AD. A novel method based on fabric phase sorptive extraction (FPSE) followed by gas chromatog.-tandem mass spectrometry (GC-MS/MS) has been validated for the simultaneous determination of 11 UV filters (ethylhexyl salicylate, benzyl salicylate, homosalate, benzophenone-3, isoamylmethoxycinnamate, 4-methylbenzylidenecamphor, Me anthranilate, etocrylene, 2-ethylhexylmethoxycinnamate, 2-ethylhexyl p-dimethylaminobenzoate, and octocrylene), in natural and recreational waters. Major exptl. parameters affecting FPSE procedure have been optimized to obtain the highest extraction efficiency. Different types and sizes of sol-gel coated FPSE media, sample volume, extraction time, and type and volume of desorption solvent were evaluated. The optimal conditions involved the use of a (2.0 x 2.5) cm2 FPSE device with PDMS based coating for the extraction of 20 mL of water for 20 min. The quant. desorption of the target compounds was performed with 0.5-1 mL of Et acetate. The method was satisfactorily validated in terms of linearity, precision, repeatability and reproducibility. Recovery studies were performed at different concentration levels in real water matrixes to show its suitability, obtaining mean values about 90% and satisfactory precision. LODs were at the low ng L-1 in all cases. Finally, the validated FPSE-GC-MS/MS method was applied to different real samples, including environmental water (lake, river, seawater) and recreational water (swimming-pool), where 8 out of the 11 studied compounds were detected at concentrations between 0.12-123μg L-1. FPSE is proposed as an efficient and simple alternative to other extraction and microextraction techniques for the anal. of UV filters in waters. Since no matrix effects were observed, quantification could be carried out by conventional calibration with standard solutions, without the need to perform the complete FPSE procedure, thus allowing a higher throughput in comparison with other microextraction techniques.
