The eye is highly susceptible to inflammation-mediated tissue damage evoked during bacterial infection. However, mechanisms regulating inflammation to protect the eye remain elusive. Here, we used integrated metabolomics and transcriptomics to show that the immunomodulatory metabolite itaconate and immune-responsive gene 1 (Irg1) are induced in bacterial (Staphylococcus aureus)-infected mouse eyes, bone-marrow-derived macrophages (BMDMs), and Muller glia. Itaconate levels are also elevated in the vitreous of patients with bacterial endophthalmitis. Irg1 deficiency in mice led to increased ocular pathol. Conversely, intraocular administration of itaconate protects both Irg1-/- and wild-type mice from bacterial endophthalmitis by reducing inflammation, bacterial burden, and preserving retinal architecture and visual function. Notably, itaconate exerts synergistic effects with antibiotics. The protective, anti-inflammatory effects of itaconate are mediated via activation of NRF2/HO-1 signaling and inhibition of NLRP3 inflammasome. Collectively, our study demonstrates the Irg1/itaconate axis is a regulator of intraocular inflammation and provides evidence for using itaconate, along with antibiotics, to treat bacterial infections.

However, one of the main disadvantages of bio-based UPR is their relatively high viscosity compared to petrochem. ones. In order to overcome this drawback, in this work, we investigated the possibility to lower the resin viscosity utilizing a mixture of di-Me itaconate (DMI) and Me methacrylate (MMA) as a reactive diluent. The effect of the DMI and MMA ratio on resin rheol. properties was investigated. The optimal curing parameters were determined and all UPRs had a high gel content, which was shown to be dependent on the DMI and MMA ratio in the formulation. Furthermore, thermomech. and mech. properties of the resulting network were also found to be affected by the used reactive diluent mixture A small substitution of DMI by MMA proved to be advantageous since it offers lower resin viscosity and improved mech. properties.

Structural advantages accrue from the increased surface area provided by the extra internal surface. Extreme sensitivity to pH and ionic strength due to ionizable moieties in the polymer network differentiates these soft colloidal particles from their uncharged counterparts, which sustain a hollow structure only at cross-link densities sufficiently high that stimuli sensitivity is reduced. Using small-angle neutron and light scattering, increased swelling of the network in the charged state accompanied by an expanded internal cavity is observed Upon addition of salt, the external fuzziness of the microgel surface diminishes while the internal fuzziness grows. These structural changes are interpreted via Poisson-Boltzmann theory in the cell model.

Immune-responsive gene 1 (irg1) is a gene that is well-conserved among different taxa and is highly expressed in the mussel Mytilus galloprovincialis at the constitutive level. The expression of this gene increases after a bacterial infection, primarily in haemocytes. Irg1 catalyzes the production of itaconic acid from cis-aconitic acid in the Krebs cycle. Recently, itaconate has been revealed as an immune metabolite involved in macrophage polarization. In this work, we studied the effects of exogenous di-Me itaconate (DI) on mussels in vitro and in vivo at relevant previously described endogenous concentrations and in mussels infected with Vibrio splendidus. DI did not have adverse effects on the haemocytes viability, apoptotic cells, proliferation and phagocytic activity; however, haemocyte size, velocity and accumulated distance were decreased. The antibacterial activity of DI in vitro and in vivo was observed with high concentrations of DI, i.e., 30 and 50 mM, resp. Furthermore, DI inhibited total ROS, increased mitochondrial ROS and modulated antioxidant genes, such as SOD and CAT, related to Nrf2 activation. In this research, we have demonstrated some important pathways in haemocytes in which itaconate can be involved after its production in a bacterial infection.

In this study, new benzoxazole-linked pyrrolidinone heterocyclic compounds I [R = H, 2-Me, 3-Cl-4-F, etc.] were synthesized by an eco-efficient strategy using substituted benzylamines and 2-aminophenol under ultrasonic irradiation in the presence of a newly designed metal-free deep eutectic solvent (DES). This DES was prepared by using a eutectic mixture of urea and a synthesized glycine-derived ionic liquid X-ray diffraction and IR spectroscopy were employed to investigate the structure of the ionic liquid and characterize the DES, resp. This method exhibited key advantages of high productivity, a short reaction time, and simple processing. Moreover, this DES was easily separated from reaction mixtures and could be recycled for multiple reactions.

The obtained pyrrolidinone derivatives I were further reacted with 2-aminothiophenol under microwave irradiation with the assistance of a deep eutectic mixture (CuCl2 or NiCl2/urea) and Cu- or Ni-doped TiO2 nanoparticles to afford the final derivatives II incorporating both pyrrolidin-2-one and benzothiazole rings. To determine the optimum reaction conditions, the effects of temperature, solvent nature, and reaction time were investigated. Under the optimal operation conditions, high reaction yields were achieved. The developed strategy exhibited advantages of a short reaction time, eco-friendliness, and high productivity. Finally, the obtained derivatives II were examined in terms of their antibacterial potential against selected Gram-neg. and Gram-pos. bacterial strains: all the derivatives II, except for II (R = 4-MeOC6H4), exhibited activity against the tested bacteria.
