A simple method is established to synthesize bio-based cross-linked polyitaconamides (cPITAs) through a Michael ene-amine addition and bulk polycondensation. A tetraester, i.e. tetra-Me piperazine-N,N’-bis(2-methylene butanedioate) (TMPB), was synthesized through a Michael addition of di-Me itaconate and piperazine at 90°C. Bulk polycondensation of TMPB with butanediamine or hexamethylenediamine was conducted at 130-170°C under atm. pressure, and several cPITAs with tunable properties were prepared The Michael addition and the polycondensation were monitored by FT-IR and 1H NMR spectra. The cPITA films were characterized by DSC, WAXS, TGA, dynamic mech. anal., and tensile test. cPITAs exhibited Tg ranging from 58 to 72°C, tensile strength up to 85 MPa, and strain at break from 9% to 17%.

Di-Me itaconate (DI), a promising biomass-derived intermediate, cannot be synthesized from itaconic acid (IA) in organisms due to the lack of metabolic pathways. It is necessary to develop a green and high-efficiency method to synthesis DI from IA. In this study, DI was, for the first time, produced from IA in an aqueous medium by an ionic liquid-strengthened enzymic method. We found that when pretreated by the ionic liquid, 1-butyl-3-methylimidazolium hexafluorophosphate ([bmim][PF6]), the pH tolerance range of the lipase shrunk and the hydrolysis activity of it was greatly inhibited, while its esterification activity was not. Moreover, when the [bmim][PF6]-treated immobilized Rhizomucor miehei lipase (RML, 50 mg/mL) was employed in a reaction containing IA/methanol at a molar ratio of 1:30 and methanol/water in a volume ratio of 1:10 at 30°C after 24 h, a 64.3% yield and 73.2% selectivity of DI was obtained. The results of HPLC and GC-MS revealed that in addition to the main product DI, other side products including acetic acid, propionic acid, and itaconic acid cyclolactone were also generated, which indicates that the RML-catalyzed esterification of IA and methanol has various side reactions, such as lactonization and self-decomposition of IA. Greenly and high-efficiently, we convert biomass-sourced itaconic acid to di-Me itaconate using an ionic liquid-strengthened enzymic method in aqueous media.

A series of Me nickel complexes with phosphino-naphtholate chelate ligands have been strategically designed, synthesized, and fully characterized. These catalysts showed extremely high activity toward ethylene polymerization and ethylene/unsaturated acid ester copolymerization without any activator under mild conditions. The catalytic activity for ethylene polymerization was up to 3.0 × 107 gPE molNi-1 h-1, which was the topmost among the neutral nickel catalysts previously reported. The endogenous metabolite itaconate and its cell-permeable derivative di-Me itaconate (DI) have been identified as anti-inflammatory regulators of macrophages; however, their contribution to inflammasome-mediated pyroptosis remains unknown. The present study examined the mol. mechanism of DI on NLR family pyrin domain-containing 3 (NLRP3) inflammasome assembly and NLRP3 inflammasome-dependent pyroptosis in microglia. Lipopolysaccharide (LPS) and ATP were used to induce microglia pyroptosis in vitro; this process was confirmed by TUNEL assay, lactate dehydrogenase (LDH) detection and gasdermin D (GSDMD) expression anal. The regulation of microglia polarization and inflammatory cytokine expression was assessed by immunofluorescence assays and ELISA. To investigate the associated mechanism of action, the expression levels of the nuclear factor erythroid 2-related factor 2 (Nrf-2)/heme oxygenase-1 (HO-1) pathway proteins were analyzed by western blotting. Finally, the regulatory effect of DI on autophagy and its association with inflammation was determined by western blotting. The present study demonstrated that DI administration inhibited NLRP3 assembly, LDH release and GSDMD cleavage. Cotreatment of DI with LPS and ATP facilitated the transition from M1 to M2, reduced inflammatory mediator expression and impeded NF-κB phosphorylation. In addition, DI effectively reduced reactive oxygen species production through the Nrf-2/HO-1 pathway. Moreover, DI induced cellular autophagy, whereas inhibition of autophagy with 3-methyladenine markedly reversed its inhibitory effect on NLRP3-dependent pyroptosis. Taken together, the present study suggested that DI participated in the Nrf-2/HO-1 pathway and served a key role in microglia inflammation and NLRP3 inflammasome-mediated pyroptosis via induction of autophagy.

The metabolite itaconate has both anti-inflammatory and immunomodulatory effects. However, its influence on chronic pain is unclear. Here, we demonstrated that i.p. injection of the itaconate derivative di-Me itaconate (DI) alleviated chronic pain symptoms, such as allodynia and hyperalgesia, in spinal nerve ligation (SNL) and inflammatory pain models. Moreover, i.p. DI reduced the secretion of inflammatory cytokines (i.e., interleukin-1β, tumor necrosis factor-alpha) in dorsal root ganglion (DRG), spinal cord and hind paw tissues, suppressed the activation of macrophages in the DRG and glial cells in the spinal dorsal horn and decreased the phosphorylation of extracellular signal-regulated kinase 1/2 (ERK1/2) in the DRG and spinal cord. DI boosted nuclear factor-erythroid 2 p45-related factor 2 (Nrf2) levels in the DRG and spinal cord of SNL mice. I.p. administration of the Nrf2 inhibitor ML385 abolished the analgesic effect of DI and decreased the expression of Nrf2 in the DRG and spinal cord. Similarly, administration of DI potently reversed the lipopolysaccharide (LPS)-induced inflammatory effect in microglia. Reduction of endogenous itaconate levels by pretreatment with immune-responsive gene 1 (IRG1) siRNA blocked Nrf2 expression, which impaired the analgesic and anti-inflammatory effects of DI in vitro. Therefore, our findings revealed for the first time that i.p. DI elicited anti-inflammatory effect and sustained chronic pain relief, which may be regarded as a promising therapeutic agent for chronic pain treatment.

Cellular metabolism plays a major role in the regulation of inflammation. The inflammatory macrophages undergo a wide-range of metabolic rewriting due to the production of significant amount of itaconate metabolite from cis-aconitate in the tricarboxylic acid cycle. This itaconate mol. has been recently described as a promising immunoregulator. However, its function and mode of action on macrophages and tissue repair and regeneration are yet unclear. Herein, the itaconate-derivative di-Me itaconate (DMI) suppresses the IL-23/IL-17 inflammatory axis-associated genes and promotes antioxidant nuclear factor erythroid 2-related factor 2 target genes. The poly-ε-caprolactone (PCL)/DMI nanofibers implanted in mice initially maintain inflammation by suppressing anti-inflammatory activity and particular inflammation, while at later stage promotes anti-inflammatory activity for an appropriate tissue repair. Furthermore, the PCL/DMI nanofiber patches show an excellent myocardial protection by reducing infarct area and improving ventricular function via time-dependent regulation of myocardium-associated genes. This study unveils potential DMI macrophage modulatory functions in tissue microenvironment and macrophages rewriting for proper tissue repair.

To study the influence of unsaturated carboxylate on the dispersing and flow releasing ability of slow-release polycarboxylate superplasticizer (PCE), the hydrolysis behaviors of carboxylates in sodium hydroxide solution at pH value of 12.0 was investigated, and the constants of hydrolysis rate were derived via a linear fitting. Mini slump was used to examine the variation of flow of cement pastes containing the synthesized slow releasing PCE over time, and the results were compared and analyzed to coordinate the relationship of hydrolysis behaviors of carboxylates and slow-release effectiveness of PCE. The results indicate that PCE with the substitution of acrylic acid with 33.3% Me acrylate and 25.0% di-Me itaconate showed significant slow-release effect, the initial fluidity of cement paste was 149 mm and 104 mm afterwards increased to 223 mm and 212 mm after 2 h resp. PCE with carboxylate monomer of faster hydrolysis rate exhibited quicker flow releasing property, and PCE with carboxylate monomer containing more ester grouped manifested stronger flow releasing ability, but need longer time. The flow releasing property of PCE not only depended on the hydrolysis rate and ester group amount of monomer, but also related to its reactivity of monomer to match the copolymerization system.
