Antimicrobial resistance is an increasingly problematic issue in the world and there is a present and urgent need to develop new antimicrobial therapies without drug resistance. Antibacterial polymers are less susceptible to drug resistance but they are prone to inducing serious side effects due to high pos. charge. Herein we report a peptide-grafted hyperbranched polymer which can self-assemble into unusual nanosheets with highly effective intrinsically antibacterial activity but weak pos. charges (+ 6.1 mV). The hyperbranched polymer was synthesized by sequential Michael addition-based thiol-ene and free radical mediated thiol-ene reactions, and followed by ring-opening polymerization of N-carboxyanhydrides (NCAs). The nanosheet structure was confirmed by transmission electron microscopy (TEM) and at. force microscopy (AFM) studies. Furthermore, a novel “wrapping and penetrating” antibacterial mechanism of the nanosheets was revealed by TEM and it is the key to significantly decrease the pos. charges but have a very low min. inhibitory concentration (MIC) of 16 μg mL-1 against typical Gram-pos. and Gram-neg. bacteria. Overall, our synthetic strategy demonstrates a new insight for synthesizing antibacterial nanomaterials with weak pos. charges. The development of efficacious carriers is an important long-standing challenge in gene therapy. Efficient delivery of nucleic acids into cells is of great interest in the field of cell biol. and gene therapy. A better understanding of the structure-function relationship of gene delivery vectors is also essential for the design of novel and intelligent delivery vectors, efficient in “difficult-to-transfect” cells and in vivo clin. applications. Most of the existing strategies for the synthesis of gene-delivery vectors require multiple steps and lengthy procedures. Here, we demonstrate a facile, three-component one-pot synthesis of a combinatorial library of 288 structurally diverse lipid-like mols. termed “lipidoids” via a thiolactone ring opening reaction. This strategy introduces the possibility to synthesize lipidoids with hydrophobic tails containing both unsaturated bonds and reducible disulfide groups. The whole synthesis and purification are convenient, extremely fast, and can be accomplished within a few hours. Screening of the produced lipidoids using HEK293T cells without addition of helper lipids resulted in identification of highly stable liposomes demonstrating ∼95% transfection efficiency with low toxicity. Visualization of intracellular transport pathways is crucial to investigate the internalization mechanism and understand the intracellular behavior of nanomaterials. Herein, we rationalized the design of micellar nanoparticles (NPs) for ratiometric fluorescent mapping of intracellular pH and glutathione (GSH), two essential parameters for maintaining normal cellular functions. Specifically, pH-sensitive naphthalimide-based probe (NPI) and pH-inert rhodamine B (RhB) were covalently labeled to double hydrophilic block copolymers (DHBCs) using the thiolactone chem., enabling the covalent attachment of NPI and RhB to one mol. with a redox-responsive disulfide linkage. The dually labeled DHBCs exhibited blue/orange dual emissions in acidic pH, which was further converted into green/orange dual emissions in neutral pH because of the deprotonation of NPI moieties and the sole green emission in the presence of GSH at neutral pH because of the decreased Forster resonance energy transfer efficiency between an NPI donor and an RhB acceptor as a result of GSH-mediated cleavage of disulfide bonds. These remarkable ratiometric fluorescence changes allowed for not only the simultaneous mapping of the intracellular pH and GSH but also the intracellular transport pathways of internalized NPs. Surface adhered bacterial colonies or biofilms are an important problem in medical and food industries. Bacteria use a chem. language to monitor their quorum and to express virulence factors, which eventually help them in colonization and manifestation of an infection. The LasR-LasI and RhlR-RhlI quorum-sensing (QS) systems of Pseudomonas aeruginosa control expression of virulence factors in a population d.-dependent fashion. In this study, we investigated the role of synthetic analogs to RhIR-RhII system of P. aeruginosa strains (PAO-1; wild-type and mutants JP-1, PDO-100, and JP-2) responsible for production of acyl-homoserine lactones-2; butanol homoserine lactone (AHL-2; C4-HSL). We synthesized double (QS1207) and single (QS0108) sulfur analogs against (C4-HSL; AHL-2), an autoinducer of Pseudomonas QS system. Extensive biol. investigation of these analogs suggested a growth promoting activity for these analogs in Pseudomonas controlling biofilm production and exo-protease secretion. We hypothesized that these thiolactone analogs could be potentially utilized as potent drug-delivery vehicles against biofilm-producing pathogens. As a proof of principle we conjugated the single sulfur analog QS0108 with the broad-spectrum antibiotic, ciprofloxacin (QS0108-Cip). The QS analog-antibiotic conjugate was significantly more effective at disrupting both the nascent and mature biofilms of P. aeruginosa than the free antibiotic. Biocompatible crosslinking is a key approach for developing biomedical hydrogels and scaffolds. In this report, native chem. ligation (NCL) was utilized to prepare biocompatible and biodegradable hydrogel using naturally derived poly(γ-glutamic acid) and ε-poly-lysine as the backbone without any additive and byproduct. First, thiolactone grafted poly(γ-glutamic acid) (PGA-HC) and cysteine grafted ε-poly-lysine (EPL-C) precursors were synthesized. Their structure was confirmed by NMR (NMR). After that, NCL crosslinking of PGA-HC and EPL-C precursors was triggered by simply blending their buffer solutions without any additive at room temperature, resulting in a hybrid polypeptide hydrogel. The crosslinking approach was verified by Fourier transform IR spectroscopy (FTIR) anal. The equilibrium water content, morphol., degradation rate and mech. properties of the hybrid hydrogels were characterized in detail. The results revealed the NCL hybrid hydrogels had tunable gelation time, water content and mech. properties by adjusting precursor composition Furthermore, the biocompatibility of hybrid hydrogels was confirmed by MTT assay. These characteristics provide a potential opportunity for the NCL hybrid polypeptide hydrogels as wound dressings, skin fillings, drug delivery vehicles and tissue regeneration matrixes.
