Due to its structural similarity, the α-1-hydroxyethyl-γ-butyrolactone obtained by reduction of (±)-α-acetyl-γ-butyrolactone may have a similar function in the body to γ-butyrolactone (GBL). In the work presented, biotransformation of α-acetyl-γ-butyrolactone by three Rhodotorula strains was performed obtaining enantiomerically enriched alc. The process was carried out in growing and resting cultures. We studied how both media composition and organic solvent volume affected stereoselectivity and effectiveness of biotransformation. After 2 h, the enantiomerically pure (3R, 1 S)-α-1-hydroxyethyl-γ-butyrolactone was obtained using the R. marina AM77 strain in YPG (Yeast extract-Peptone-Glucose) medium enriched with 5% glycerol. To our best knowledge there is no previous information in the literature about the (±)-α-acetyl-γ-butyrolactone biotransformation performed in medium with addition of organic and deep eutectic solvents. The authors found that the chlorination reaction can be carried out in only 30 s at room temperature by flow. The decarboxylation/acylation step was also simplified by using a cross-mixer, so that acetic anhydride was no longer required in the acylation reaction. A computational fluid dynamics simulation was carried out to study the improved micromixing of liquid-liquid two-phase streams. Finally, 3-chloro-4-oxopentyl acetate was obtained in a 90% isolated yield with a product purity of 96% and a total residence time of approx. 32 min. This fully continuous process was operated smoothly for 12 h, and approx. 19.1 g of the desired product was generated with a production rate of 1.79 g h-1. To understand the fast pyrolysis kinetics and product evolution of waste pine sawdust, high heating rate thermogravimetry-Fourier transform IR (TG-FTIR) was used to obtain the kinetic parameters and the chem. groups formed during the pyrolysis process, while pyrolysis-gas chromatog./mass spectrometry (Py-GC/MS) was used to investigate the detailed compositions of products under the staged (seven stages from 300 to 600°C) and direct fast pyrolysis process. Spectral bands were identified for acids, alcs., aldehydes, aromatics, esters, ethers, hydrocarbons, ketones, phenols, and sugars. Research found that the apparent activation energy for fast pyrolysis is much higher than that of slow pyrolysis. The evolution of CO2 is the major deoxygenation route. Cracking mainly occurred at the 450°C stage with phenols, ketones, aldehydes, and sugars as the main products. The product distributions for different stages are significantly different; the selectivity of aldehydes decreased, while phenols showed an upward trend with an increase in pyrolysis temperature Ketones and sugars reached their peak values at 450°C. The changes in the mol. composition of each stage helped to understand the pyrolysis process. Compared with the staged pyrolysis, the direct pyrolysis process had higher selectivity of acids, aldehydes, esters, and sugars and lower selectivity of phenols, ketones, and alcs. Two isosteric series of 1,2,4-triazole and 1,3,4-thiadiazole derivatives were synthesized and evaluated for their antiviral activity against herpes simplex virus type 1, HSV-1, using acyclovir (ACV) as a reference drug. In addition, mol. docking into the active site of HSV-1 thymidine kinase was performed to interpret the data obtained from biol. testing, and all compounds were subjected to an in-silico screening of their physicochem. properties to estimate their drug-likeness and safety. The results revealed that triazolopyrimidine derivative I was able to reduce the viral plaques by 50% at a dose of 80 μM, but interestingly, retained high selectivity compared to ACV (>200 μM vs. 80 μM). The best effective and safe compound in this study, triazolopyrimidine derivative I, was further tested for its combined effects with ACV on the anti-HSV-1 activity in the plaque reduction assay. This compound proved to improve the selectivity of ACV and reduce its ED that produced 100% inhibition of viral plaques. The compound I was suggested to be a promising candidate for further development as an antiherpetic agent. Mol. docking into the active site of HSV-1 thymidine kinase emphasized the superior interaction of the compound I.
