Evaluation of nutritional composition and ecotoxicity of the stick insect Cladomorphus phyllinum

The global population is estimated to reach 9 billion at 2050; thus, there will be an increase of food demand. In this scenario, edible insects can be considered as an option for both food and supplements source. In this work was evaluated the nutritional aspects of Cladomorphus phyllinum, also known as stick insect. Protein, carbohydrates, amino acids, lipids, fatty acids, minerals and ecotoxicity were analyzed. The raw sample had protein, carbohydrate, ester and lipid content of 17.96%, 8.33%, 15.23% and 1.85%, respectively, while the dry sample presented 64.6% protein content. It has been found that the dry stick insect presented the essential amino acids, leucine, isoleucine, valine, lysine, threonine, methionine, phenylalanine and histidine. Oleic acid (57.03%), palmitic acid (15.94%), linoleic acid (13.76%) and stearic acid (10.67%) were also found in dry samples. The micronutrients calcium, magnesium and zinc were the minerals with expressive concentrations. Besides, acute ecotoxicity tests performed with extremely sensitive species, Daphnia magna, Vibrio fischeri and Artemia Salina, revealed no toxicity associated with the Cladomorphus phyllinum. These results obtained in this work indicate that the stick insect can be used as an alternative insect-based protein, minerals, fatty acids and amino source.

Assessment of plastic ingestion by pole-caught pelagic predatory fish from O’ahu, Hawai’i

Although the frequency of occurrence of plastic ingestion in the large-sized dolphinfish and tunas taken by the Hawai’i longline fishery is very low (frequency of occurrence < 5% of sampled individuals), the ingestion of plastic in smaller-sized specimens caught with pole-and-line gear by commercial and recreational fishers has not been investigated. This study examined ingestion of >0.25 mm marine plastic debris (MPD) by four predatory fish species caught by commercial fishers around the Main Hawaiian Islands, and documented ingestion in three species: 85.7% of albacore tuna (n = 7), 40.0% of skipjack tuna (n = 10) and 12.5% of dolphinfish (n = 8). Yellowfin tuna (n = 10) did not contain any MPD, probably owing to the high proportion of empty stomachs (60%). For skipjack tuna, the frequency of occurrence of MPD ingestion was significantly higher for the smaller-sized specimens caught with pole-and-line (40%), compared with the larger-sized specimens caught with longlines (0%). For dolphinfish, the frequency of occurrence of MPD ingestion was similar for the similar-sized specimens caught with pole-and-line and with longlines. The ingested MPD items were micro-meso plastics, between 1 and 25 mm. While most ingested items were fragments, albacore also ingested line and skipjack also ingested sheets. The predatory fishes ingested light MPD items that float in sea water, but there were species-specific differences in their polymer composition: albacore contained more polypropylene and polyethylene, and skipjack contained more elastomers, characterized by a high percentage of ester plasticizers. Altogether, these results suggest that albacore and skipjack tunas ingest plastic of different types and polymers. Yet more research is needed to understand how differences in vertical distribution, foraging ecology and diet influence the MPD sampled by these predatory fish species.

Floral traits and chemical cues associated with rock bee (Apis dorsata Fabricius) for the host selection in West Bengal, India

In the present work, bee foraging plants were determined by palynological analysis of colony stored samples (honey and pollen) of Apis dorsata collected from southern West Bengal, India. Then flowering phenology and pollen morphologies were described to evaluate the forage preference of the bee species. In addition, chemical cues were analysed using gas chromatography-mass spectrometry (GC-MS). Foraging strategy of the bee species was profitable in that almost all the bee-visited plants were utilised for both nectar and pollen sources. The bee species became generalist visitor of several plants having diverse life form, flower characters and pollen morphologies. However, most of the bee-visited plants were of trees with synchronous type of flowering. Most plants had small-sized, yellow, cream and white coloured flowers and pollen was openly presented. We did not find any patterns in the morphological traits of bee collected pollen, though the pollen types were predominantly of medium sized with trizonocolporate apertural pattern and reticulate surface ornamentation. Floral volatile organic compounds (VOCs) of bee visited plants were also diversified. Compounds frequently found in the floral VOC bouquets were amphetamine-3-methyl; butane, 2-cyclopropyl; 2,3-butanediol; cyclohexan, 1-methyl-5-(1-methylethyl); d-limonene; methyl (2E)-2-methoxy-2-butenoate; phenol, 4-[2-(methylamino)ethyl]; phthalic acid, di(2-propylpentyl)ester; propanamide, N-(aminocarbonyl) and pyrrolo[1,2-a]pyrazine-1,4-dione, hexahydro-3-(2-methylpropyl). Metabolism and pharmacokinetics of a novel polyphenol fatty acid ester phloridzin docosahexaenoate in Balb/c female mice

Flavonoids are known to undergo phase II metabolism and produce metabolites with similar or stronger biological effects compared to the parent flavonoids. However, the limited cellular uptake and bioavailability restrict their clinical use. We synthesized phloridzin docosahexaenoate (PZ-DHA), a novel fatty acid ester of polyphenol, through an acylation reaction with the aim of increasing the cellular availability and stability of the parent biomolecules, phloridzin (PZ) and docosahexaenoic acid (DHA). Here, we report metabolites and pharmacokinetic parameters of PZ-DHA, determined using ultra-high-performance liquid chromatography-electrospray ionization tandem mass spectrometry. PZ-DHA was taken-up by human (MDA-MB-231, MDA-MB-468, and MCF-7) and mouse (4T1) mammary carcinoma and human non-malignant mammary epithelial cells (MCF-10A) in cellular uptake assays. Our results suggested that the acylation improves the cellular uptake of PZ and stability of DHA within cells. In mouse hepatic microsomal assays, two major glucuronides of PZ-DHA, PZ-DHA-4-O-glucuronide and PZ-DHA-4 ‘-O-glucuronide (MW=923.02 g/mol), were detected. One tri-methylated- (4,4 ‘,6 ‘-O-trimethyl-PZ-DHA) (MW=788.88 g/mol) and one di-sulphated- (PZ-DHA-4,4 ‘-O-disulphide) PZ-DHA metabolite (MW=906.20 g/mol) were also identified. Intraperitoneal injections of PZ-DHA (100 mg/kg) into Balb/c female mice was rapidly absorbed with a serum C-max and T-max of 23.7 mu M and 60 min, respectively, and rapidly eliminated (t(1/2)=28.7 min). PZ-DHA and its metabolites are readily distributed throughout the body (V-d=57 mL) into many organs. We identified in vitro and in vivo metabolites of PZ-DHA, which could be tested for potential use to treat diseases such as cancer in multiple organ systems.

Crude glycerol impurities improve Rhizomucor miehei lipase production by Pichia pastoris

Crude glycerol, a by-product of biodiesel production, was employed as the carbon source to produce lipase using Pichia pastoris. Under identical fermentation conditions, cell growth and lipase activity were improved using crude glycerol instead of pure glycerol. The impacts of crude glycerol impurities (methyl ester, grease, glycerol, methanol, and metal ions Na+, Ca2+, and Fe3+) on lipase production were investigated. Impurities accelerated P. pastoris entering the stationary phase. Na+, Ca2+, and grease in waste crude glycerol were the main factors influencing higher lipase activity. Through response surface optimization of Ca2+, Na+, and grease concentrations, lipase activity reached 1437 U/mL (15,977 U/mg), which was 2.5 times that of the control. This study highlights the economical and highly efficient valorization of crude glycerol, demonstrating its possible utilization as a carbon source to produce lipase by P. pastoris without pretreatment.
