Free fatty acid receptor 3 (FFA3, previously GPR41) is activated by short-chain fatty acids, mediates health effects of the gut microbiota, and is a therapeutic target for metabolic and inflammatory diseases. The shortage of well-characterized tool compounds has however impeded progress. Herein, we report structure-activity relationship of an allosteric modulator series and characterization of physicochem. and pharmacokinetic properties of selected compounds, including previous and new tools. Two representatives, 57 (TUG-1907) and 63 (TUG-2015), showed improved solubility and preserved potency. Of these, 57, with EC50 = 145 nM and a solubility of 33μM, showed high clearance in vivo but is a preferred tool in vitro. In contrast, 63, with EC50 = 162 nM and a solubility of 9μM, showed lower clearance and seems better suited for in vivo studies. Using 57, we demonstrate for the first time that FFA3 activation leads to calcium mobilization in murine dorsal root ganglia. 《Organocatalytic Atroposelective Synthesis of N-N Axially Chiral Indoles and Pyrroles by De Novo Ring Formation》 was written by Chen, Ke-Wei; Chen, Zhi-Han; Yang, Shuang; Wu, Shu-Fang; Zhang, Yu-Chen; Shi, Feng. Application of 30414-53-0 And the article was included in Angewandte Chemie, International Edition on April 19 ,2022. The article conveys some information:

The first highly atroposelective construction of N-N axially chiral indole scaffolds I [R = H, 4-F, 6-MeO, 7-Br, etc.; R1 = H, Me; R2 = CO2Me, CO2Et, Ph, etc.; R3 = Me, Et, i-Pr, Bn; R4 = Ph, 2-FC6H4, 3-FC6H4, etc.] was established via a new strategy of de novo ring formation. This strategy made use of the organocatalytic asym. Paal-Knorr reaction of well-designed N-aminoindoles with 1,4-diketones, thus affording N-pyrrolylindoles in high yields and with excellent atroposelectivities (up to 98% yield, 96% ee). In addition, this strategy was applicable for the atroposelective synthesis of N-N axially chiral bispyrroles II [R5 = Ph, 3-ClC6H4, 3-MeOC6H4, 1-naphthyl, 2-naphthyl; R6 = Me, Et, Bn; R7 = Me, Et, i-Pr; R8 = Ph, 2-FC6H4, 3-thienyl, etc.; R9 = Me, Et, i-Pr, Bn] (up to 98% yield, 97% ee). More importantly, such N-N axially chiral heterocycles could be converted into chiral organocatalysts with applications in asym. catalysis, and some mols. display potent anticancer activity. This work not only provided a new strategy for the atroposelective synthesis of N-N axially chiral mols. but also offers new members of the N-N atropisomer family with promising applications in synthetic and medicinal chem. The information in the text is summarized as follows:

The biocatalytic stereocontrolled synthesis of various acyclic anti- or syn-α-alkyl-β-amino amides through a dynamic kinetic resolution strategy is demonstrated. A series of com. available and inhouse transaminases (TAs) were employed to perform the transamination of a series of chem. synthesized racemic α-alkyl-β-keto amides. Among them, com. (R)-selective TAs showed the best activities and selectivities, giving access preferentially to the anti-diastereoisomers with low to high diastereomeric ratios (up to 96%) and excellent enantiomeric excess (>99%). The stereoselective biotransamination experiments were successfully demonstrated at a semi-preparative scale (25 mM, 100 mg substrate), leading to the corresponding optically active α-alkyl-β-amino amides in 45-90% isolated yields after a simple liquid-liquid extraction protocol. Chiral (α-substituted) β-hydroxy amides are interesting derivatives as they are useful building blocks of many biol. active compounds Herein, the biocatalytic stereocontrolled synthesis of various acyclic syn-α-alkyl-β-hydroxy amides through a dynamic kinetic resolution is shown. The article was written by Wang, Xiao-Mei; Zhang, Peng; Xu, Qi; Guo, Chang-Qiu; Zhang, De-Bing; Lu, Chuan-Jun; Liu, Ren-Rong. The article contains the following contents:

Nitrogen-nitrogen bonds containing motifs are ubiquitous in natural products and bioactive compounds However, the atropisomerism arising from a restricted rotation around an N-N bond is largely overlooked. Here, a method to access the first enantioselective synthesis of N-N biaryl atropisomers via a Cu-bisoxazoline-catalyzed Friedel-Crafts alkylation reaction is described. A wide range of axially chiral N-N bisazaheterocycle compounds were efficiently prepared in high yields with excellent enantioselectivities via desymmetrization and kinetic resolution Heating experiments showed that the axially chiral bisazaheterocycle products have high rotational barriers. The article was titled 《Molecular Engineering of β-Substituted Oxoporphyrinogens for Hydrogen-Bond Donor Catalysis》, and you may find the article in European Journal of Organic Chemistry.Application of 30414-53-0 The information in the text is summarized as follows:

A new class of bifunctional hydrogen-bond donor organocatalyst using oxoporphyrinogens having increased intramol. hydrogen-bond donor distances is reported. Oxoporphyrinogens are highly non-planar rigid macrocycles containing a multiple hydrogen bond-forming binding site. In this work, we describe the first example of non-planar OxPs as hydrogen-bond donor catalysts prepared using a mol. engineering approach of the binding site for dual activation of substrates. The introduction of β-substituents is key to the catalytic activity and the catalysts are able to catalyze 1,4-conjugate additions and sulfa-Michael additions, as well as, Henry and aza-Henry reactions at low catalyst loadings (≤ 1 mol-%) under mild conditions (e.g., catalyst I). Preliminary mechanistic studies have been carried out and a possible reaction mechanism has been proposed based on the bi-functional activation of both substrates through hydrogen-bonding interactions. A novel five-component stereoselective synthesis of polysubstituted tetrahydropyridines is reported. The Knoevenagel condensation – Michael addition – Mannich reaction – cyclization – dehydration cascade of aldehydes, esters of 3-oxocarboxylic acids C-H acids and ammonium acetate provides convenient access to 2-substituted alkyl (4SR,6RS)-4,6-diaryl-5,5-dicyano-1,4,5,6-tetrahydropyridine-3-carboxylates with two stereocenters and 3,5-dialkyl (4RS,5SR,6RS)-5-cyano-2,4,6-triaryl-1,4,5,6-tetrahydropyridine-5,3-carboxylates with three stereocenters in 57-84% yields. It was established that formation of products proceeds via substituted 2-hydroxypiperidines with four stereocenters. Ammonium acetate plays a dual role, acting as a base and as a nitrogen source. Five bonds are formed as a result of multicomponent process. The formation of single diastereomers was confirmed by singe crystal X-ray diffraction studies and 2D-NMR spectroscopy.
