Kunishige, Rina published the artcileCalciprotein particle-induced cytotoxicity via lysosomal dysfunction and altered cholesterol distribution in renal epithelial HK-2 cells, Quality Control of 2044-85-1, the main research area is calciprotein cholesterol distribution lysosomal dysfunction cytotoxicity.

Dietary phosphate overload induces chronic kidney disease (CKD), and calciprotein particles (CPPs), a form of nanoparticle comprising calcium phosphate and serum proteins, has been proposed to cause renal toxicity. However, the mechanism of CPP cytotoxicity in renal tubular cells is unknown. Here we show that in renal proximal tubular epithelial HK-2 cells, endocytosed CPPs accumulate in late endosomes/lysosomes (LELs) and increase their luminal pH by ∼ 1.0 unit. This results in a decrease in lysosomal hydrolase activity and autophagic flux blockage without lysosomal rupture and reactive oxygen species generation. CPP treatment led to vulnerability to H2O2-induced oxidative stress and plasma membrane injury, probably because of autophagic flux blockage and decreased plasma membrane cholesterol, resp. CPP-induced disruption of lysosomal homeostasis, autophagy flux and plasma membrane integrity might trigger a vicious cycle, leading to progressive nephron loss.

Li, Jie-xia published the artcileDoxorubicin-loaded hydrogen peroxide self-providing copper nanodots for combination of chemotherapy and acid-induced chemodynamic therapy against breast cancer, Product Details of C24H14Cl2O7, the main research area is doxorubicin hydrogen peroxide copper nanodot chemodynamic therapy breast cancer; Acid induced; Chemical dynamic therapy (CDT); Copper nanodots; H(2)O(2) self-providing; Oxygen-independent.

In recent years, chemodynamic therapy (CDT) has gained increasing interest in cancer treatment. In contrast to photodynamic therapy and sonodynamic therapy, extrinsic excitations such as laser or ultrasound are not required in CDT. As a result, the CDT performance is not limited by the penetration depth of the external irritation. However, CDT relies heavily on hydrogen peroxide (H2O2) in the tumor microenvironment (TME). Insufficient H2O2 in the TME limits the CDT performance, and the most reported methods to produce H2O2 in the TME are dependent on oxygen supply, which is restricted by the hypoxic TME. In this study, H2O2 self-providing copper nanodots were proposed, and the drug doxorubicin (DOX) was successfully loaded to construct DOX-nanodots. Our results showed that the nanodots produced H2O2 in the weakly acidic TME due to the peroxo group and further generated the most active hydroxyl radical (·OH) through the Fenton-like reaction. This process was pH-dependent and did not occur in a neutral environment. In addition to ·OH, the nanodots also produced singlet oxygen (1O2) and superoxide anions (O-2) in the cancer cells. The copper nanodots performed promising CDT against breast cancer in vitro and in vivo, with enhanced cell apoptosis and decreased cell proliferation. The combination of chemotherapy and CDT using DOX-nanodots further improved the therapeutic effects. The treatments showed good biocompatibility with no obvious toxicity in major tissues, possibly due to the specific ·OH generation in the weakly acidic TME. In summary, the H2O2 self-providing copper nanodots in combination with DOX showed promising cancer-curing effects due to the oxygen-independent and tumor-specific production of reactive oxygen species and the cooperation of chemotherapy.

Hyun, Soo-Wang published the artcileToxicological effects of urban particulate matter on corneal and conjunctival epithelial cells, Recommanded Product: 2′,7′-Dichloro-3-oxo-3H-spiro[isobenzofuran-1,9′-xanthene]-3′,6′-diyl diacetate, the main research area is toxicol effect corneal conjunctival epithelial cell; Cell cycle; Cell death; Corneal/conjunctival epithelial cells; Mucin; Urban particulate mater.

Abstract: Exposure to urban particulate matter (UPM) is a high-risk factor for various ocular surface diseases, including dry eye syndrome. However, the effects of UPM on corneal and conjunctival epithelium damage have not been fully elucidated. In this study, we investigated the toxicol. effects of UPM exposure at high concentrations by using in vitro cultures. The cell viability, mucin expression, and the secreted inflammatory mediators of corneal and conjunctival epithelial cells was observed at 24 h after exposure to UPM. The progression of cell cycle was also examined by flow cytometry at 24 h after exposure to UPM. UPM reduced cell viability in a dose-dependent manner and increased cell population in S and G2 phase. The expression of mucin-1 was attenuated by UPM exposure, but that of mucin-4 was not. UPM increased interleukin (IL)-6 release and decreased IL-8 release. The intensity of 2′ ,7′-dichlorofluorescein diacetate (DCF-DA) was highest at 4 h of UPM exposure. In conclusion, these results suggest that UPM causes the disruption of corneal and conjunctival epithelium by decreasing cell viability, altering cell cycle, disrupting mucin, and regulating inflammatory mediators.

In this work, dual-emissive ratiometric fluorescent system was constructed by the introduction of an ideal internal reference By virtue of its unique alkalinity, N2H4 could undergo a hydrazinolysis reaction with the ester group of F1, inducing remarkable fluorescence enhancement while the blue fluorescence of the internal reference DPA remained constant Consequently, the fluorescence intensity ratios (I540/I440) were proportional to the concentrations of N2H4, which was beneficial for the exactly quant. detection. The skillful strategy granted the sensing system advantages such as relative good solubility in aqueous media, easy-to-design, simple synthesis, large emission shift, good ratiometric response, as well as the successful application in real water samples and cell imaging.

Chemical Research in Toxicology published new progress about Adsorption. 2044-85-1 belongs to class esters-buliding-blocks, name is 2′,7′-Dichloro-3-oxo-3H-spiro[isobenzofuran-1,9′-xanthene]-3′,6′-diyl diacetate, and the molecular formula is C24H14Cl2O7, Product Details of C24H14Cl2O7.

Kessler, Amanda published the artcileAdsorption of Horseradish Peroxidase on Metallic Nanoparticles: Effects on Reactive Oxygen Species Detection Using 2′,7′-Dichlorofluorescin Diacetate, Product Details of C24H14Cl2O7, the main research area is adsorption horseradish peroxidase metallic nanoparticle fluorescence indicator; reactive oxygen species dichlorofluorescin diacetate.

The fluorescent probe 2′,7′-dichlorofluorescein diacetate (DCFH-DA) together with the enzyme horseradish peroxidase (HRP) is widely used in nanotoxicol. to study acellular reactive oxygen species (ROS) production from nanoparticles (NPs). This study examined whether HRP adsorbs onto NPs of Mn, Ni, and Cu and if this surface process influences the extent of metal release and hence the ROS production measurements using the DCFH assay in phosphate buffered saline (PBS), saline, or Dulbecco’s modified Eagle’s medium (DMEM). Adsorption of HRP was evident onto all NPs and conditions, except for Mn NPs in PBS. The presence of HRP resulted in an increased release of copper from the Cu NPs in PBS and reduced levels of nickel from the Ni NPs in saline. Both metal ions in solution and the adsorption of HRP onto the NPs can change the activity of HRP and thus influence the ROS results. The effect of HRP on the NP reactivity was shown to be solution chem. dependent. Most notable was the evident affinity/adsorption of phosphate toward the metal NPs, followed by a reduced adsorption of HRP, the concomitant reduction in released manganese from the Mn NPs, and increased levels of released metals from the Cu NPs in PBS. Minor effects were observed for the Ni NPs. The solution pH should be monitored since the release of metals can change the solution pH and the activity of HRP is known to be pH-dependent. It is furthermore essential that solution pH adjustments are made following the addition of NaOH during diacetyl removal of DCFH-DA. Even though not observed for the given exposure conditions of this study, released metal ions could possibly induce agglomeration or partial denaturation of HRP, which in turn could result in steric hindrance for H2O2 to reach the active site of HRP. This study further emphasizes the influence of HRP on the background kinetics, its solution dependence, and effects on measured ROS signals. Different ways of correcting for the background are highlighted, as this can result in different interpretations of generated results. The results show that adsorption of HRP onto the metal NPs influenced the extent of metal release and may, depending on the investigated system, result in either under- or overestimated ROS signals if used together with the DCFH assay. HRP should hence be used with caution when measuring ROS in the presence of reactive metallic NPs.

Fabricating antibacterial hydrogels with antimicrobial drugs and synthetic biocompatible biomimetic hydrogels is a promising strategy for practical medical applications. Here, we report a bicomponent hydrogel composed of a biomimetic polyisocyanopetide (PIC) hydrogel and a photodynamic antibacterial membrane-intercalating conjugated oligoelectrolyte (COE). The aggregation behavior and aggregate size of the COEs in water can be regulated using the PIC hydrogel, which could induce COEs with higher reactive oxygen species (ROS) production efficiency and increased association of COEs toward bacteria, therefore enhancing the antibacterial efficiency. This strategy provides a facile method for developing biomimetic hydrogels with high antibacterial capability.

Samantaray, Paresh Kumar published the artcile‘Template-free’ hierarchical MoS2 foam as a sustainable ‘green’ scavenger of heavy metals and bacteria in point of use water purification, Category: esters-buliding-blocks, the main research area is molybdenum sulfide heavy metal bacteria.

Molybdenum disulfide (MoS2), with its unique optical and elec. properties, has been explored for a variety of applications in the recent past. Still, its capabilities in point-of-use heavy metal ion removal remain to be explored. Herein, for the first time using a facile approach, we fabricated three-dimensional (3D) MoS2 foam from exfoliated single to few-layered MoS2 sheets for the selective exclusion of heavy metals and stringent bactericidal response. This foam was able to exclude 99.9% of Pb(II) and 98.7% of As(III) instantaneously and reduced more than 98% of bacteria E. coli. Moreover, the foam exhibits selective toxicity towards bacterial cells while showing no observable toxicity towards mammalian cells. The foam can be recycled and reused for at least five cycles under accelerated conditions and thus can be used for a promising non-cytotoxic, facile, and environmentally benign process for inline water remediation to remove heavy metal ions from the feed and as a potential antibacterial agent.

Liang, Zhihao published the artcileDose-Dependent Effect of ZnO Quantum Dots for Lettuce Growth, COA of Formula: C24H14Cl2O7, the main research area is zinc oxide quantum dot lettuce biomass antioxidant.

As the cadmium-free semiconductor quantum dots, ZnO quantum dots (ZnO QDs) have wide potential applications in agriculture. However, the effects of ZnO quantum dots on crop growth and nutritional quality have not been fully studied. In this work, the lettuce was sprayed with different concentrations of ZnO QDs from 50 to 500 mg·L-1 to evaluate their influence on lettuce antioxidant, biomass, and nutritional quality. The results showed that ZnO QDs existed in the lettuce in the form of Zn2+. Lettuce treated with 500 mg·L-1 ZnO QDs would produce a large amount of reactive oxygen species (ROS), which adversely affected the absorption of nutrients, soluble protein content, and chlorophyll content, thus reducing plant biomass. When the concentrations range from 50 to 200 mg·L-1, the antioxidant enzyme systems of lettuce were triggered to counteract the damage caused by excessive ROS. Moreover, ZnO QDs at this level promoted Ca, Mg, Fe, Mn, Zn, and B absorption and accumulation; increased soluble sugar content; and improved the lettuce biomass and nutritional quality.

The bacteria redox balance between oxidizing and reducing species plays a critical role in bacterial activities, and the disruption of this homeostasis offers a flexible antibacterial strategy to combat bacterial multidrug resistance. Here, the ligand exchange strategy of Au NCs was first developed to construct an oxidative stress amplifier. We cleverly utilized the reactive oxygen species (ROS) generation ability of histidine (His)-stabilized Au NCs. Cinnamaldehyde (CA) was modified on the surface of Au NCs through an aldimine condensation reaction, and the modification of CA on the surface of Au NCs further accelerated ROS generation. Meanwhile, the strong Au-S interaction between CA-Au NCs and thiols facilitated the ligand exchange of surface histidine-cinnamaldehyde (His-CA) with thiol mols., causing the consumption of thiols in bacteria and the release of His-CA, which thus finally resulted in efficient bacterial cell death. CA-Au NCs showed excellent antibacterial effects on methicillin-resistant Staphylococcus aureus (MRSA), including 48-h biofilm removal and the treatment of a pig skin wound infection model, representing a promising antibacterial agent for clin. applications.
