The volatile composition of dichloromethane extracts of preputial glandular secretions (PGS) from intact and castrated male Brandt’s voles Lasiopodomys brandtii were analyzed by using gas chromatog.-mass spectrometry (GC-MS). Thirty-three volatile compounds were detected and were present in nearly all scent donors, and 27 had previously been reported as insect pheromone components. Castration did not eliminate any compounds; however, it significantly suppressed the relative quantity of ten early-eluting, low mol. weight components. These included seven saturated and unsaturated acetates, one hexanoate, and two saturated octanoates. In particular, E,E-farnesyl acetate (FA) was the most abundant compound in the PGS. FA and other components might be candidates for male-produced pheromones. Two-choice behavioral assays of females revealed that low concentrations of raw PGS (0.056%) and FA (5 ppm in water v/w) were attractive, whereas high concentrations (0.56% and 50 ppm, resp.) were repellent. This suggested that FA might act as a male pheromone in a dosage-dependent manner. Extremely high variability of the relative quantities of almost all detected compounds among individuals indicated the inter-individual dissimilarities of PGS and consequent possibilities to communicate individuality to other conspecific members.

Sex pheromones of the fall armyworm, Spodoptera frugiperda, show differences in composition and proportions in different geog. populations, but always contain Z9-14:OAc as the major component. Odorant receptor neurons (ORNs) housed in the long trichoid sensilla (TS) of male antennae are essential to detect female-released sex pheromones in moths. In this study, we identified seven components from pheromone gland extracts of female S. frugiperda in the Yunnan population from China, including (Z)-7-dodecen-1-yl acetate (Z7-12:OAc), (Z)-9-tetradecenal (Z9-14:Ald), (Z)-9-dodecen-1-yl acetate (Z9-12:OAc), (Z)-9-tetradecen-1-yl acetate (Z9-14:OAc), (E)-11-tetradecen-1-yl acetate (E11-14:OAc), (Z)-11-tetradecen-1-yl acetate (Z11-14:OAc) and (Z)-11-hexadecen-1-yl acetate (Z11-16:OAc) at a ratio of 1.2:4:0.8:79.1:1.6:1.6:11.7 by gas chromatog. coupled with mass spectrometry. Gas chromatog.-electroantennog. detection showed that Z9-14:OAc, Z7-12:OAc and Z11-16:OAc are the male antennal active components. Peripheral coding of pheromones in males was investigated by single sensillum recording. Five functional neurons housed in three types of TS were identified based on profiles of neuronal responses, which are responsible for attractive component Z9-14:OAc, synergistic components Z7-12:OAc, Z11-16:OAc, interspecific pheromones (Z)-9-tetradecen-1-ol (Z9-14:OH) and (Z,E)-9,12-tetradecadien-1-yl acetate (Z9,E12-14:OAc), resp. Wind tunnel and field tests demonstrated that a ternary combination of Z9-14:OAc, Z7-12:OAc and Z11-16:OAc at a ratio of 88:1:11 shows the strongest attractiveness to males. Trapping tests in Niger showed that the major component, Z7-12:OH, and two of the minor components, Z5-10:OH and Z7-12:Ald, were essential for attraction of male C. ignefusalis moths. The most attractive blend contained these three components in a 100:5:3.3 ratio in a polyethylene vial, which emitted the components in similar proportions to those produced by the female C. ignefusalis moth. Water traps baited with this blend containing 1 mg of Z7-12:OH caught more male C. ignefusalis moths than traps baited with newly emerged female moths. Addition of up to 10% of the corresponding E isomers of the pheromone components had no effect on catches, but addition of the other two minor components detected, Z7-12:Ac and/or Z9-14:OH, to the attractive blend at naturally occurring levels caused significant reductions in trap catch.

The European grapevine moth, Lobesia botrana, uses (E,Z)-7,9-dodecadienyl acetate as its major sex pheromone component. Through in vivo labeling experiments we demonstrated that the doubly unsaturated pheromone component is produced by Δ11 desaturation of tetradecanoic acid, followed by chain shortening of (Z)-11-tetradecenoic acid to (Z)-9-dodecenoic acid, and subsequently introduction of the second double bond by an unknown Δ7 desaturase, before final reduction and acetylation. By sequencing and analyzing the transcriptome of female pheromone glands of L. botrana, we obtained 41 candidate genes that may be involved in sex pheromone production, including the genes encoding 17 fatty acyl desaturases, 13 fatty acyl reductases, 1 fatty acid synthase, 3 acyl-CoA oxidases, 1 acetyl-CoA carboxylase, 4 fatty acid transport proteins and 2 acyl-CoA binding proteins. A functional assay of desaturase and acyl-CoA oxidase gene candidates in yeast and insect cell (Sf9) heterologous expression systems revealed that Lbo_PPTQ encodes a Δ11 desaturase producing (Z)-11-tetradecenoic acid from tetradecanoic acid. Further, Lbo_31670 and Lbo_49602 encode two acyl-CoA oxidases that may produce (Z)-9-dodecenoic acid by chain shortening (Z)-11-tetradecenoic acid. The gene encoding the enzyme introducing the E7 double bond into (Z)-9-dodecenoic acid remains elusive even though we assayed 17 candidate desaturases in the two heterologous systems.

Spodoptera frugiperda among the China population employs a four-component sex pheromone blend to accelerate male-female allocation and mating behavior. The underlying mol. mechanism has been incompletely elucidated. In the current study, we showed that differences existed between genders toward the four sex pheromone components, including Z9-14:AC, Z7-12:AC, Z9-12:AC, and Z11-16:AC, in terms of electrophysiol. responses and behavioral valences. Male adults were significantly more sensitive to all tested compounds than female adults. Furthermore, ecol. outputs may be related to four pheromone-binding proteins, namely, SfruPBP1, SfruPBP2, SfruPBP3, and SfruPBP4. They formed four distinct clades within the lepidopteran phylogeny, and male adults expressed significantly higher levels of SfruPBP1 and SfruPBP2 than female adults. We observed the highest binding affinities of SfruPBP1 toward all four sex pheromone components. SfruPBP4 had moderate binding affinities for Z7-12:AC, Z11-16:AC, and Z9-12:AC, while SfruPBP2 showed binding toward Z9-14:AC. This observation suggests that SfruPBP1 plays a key role in sex pheromone discrimination and drives sexually biased behavioral decisions toward certain pheromone components. These findings will help to develop behavioral-mediating tools as part of integrated pest management approaches for this cross-border pest.
