Food additives are used in numerous food products and are characterised by various physicochem. properties. In European member states, their use in food is regulated by the European Union. This work aimed to develop an accurate and high-throughput anal. method enabling the simultaneous determination of additives from different functional classes to facilitate controls and generate occurrence data for exposure assessments. The QuEChERS principle was applied due to its ease of implementation and flexibility to adjust to various food matrixes. However, very polar substances could not be extracted with sufficient recoveries. Consequently, an alternative basic methanol sample-preparation methodol. was developed. After sample preparation, the obtained extracts were analyzed using ultra-high-performance liquid chromatog. coupled to tandem mass spectrometry (UHPLC-MS/MS). Overall, the developed methodol. allowed the quantification of 27 additives from the functional classes of colors, sweeteners, preservatives, and antioxidants in various foods (e.g. beverages, dairies, processed meals). The methods were also validated in terms of selectivity, linearity, matrix effect, limit of quantification, accuracy, repeatability, and intra-laboratory reproducibility. Finally, the methods were successfully applied to eighty-four actual samples. All additives were found below authorised levels. However, irregularities were spotted in labeling.

Synthetic phenolic antioxidants (SPAs) are widely used as food additives to delay the oxidation rate of oils and oil products. The concentrations and compositions of SPAs in Chinese residents most popular daily foods and the resulting exposure risk of SPAs are not clear. Therefore, this study collected food samples in 13 food categories (n = 289) from 10 provinces in China. At least one of the SPAs was detected in approx. 99.7% of foodstuffs, and the concentration of ∑SPAs ranged from not detected to 7830 (geometric mean (GM): 296 ng/g wet weight). 2,6-2 tert Bu p-1,4-benzoquinone (BHT-Q) was the main transformation product, but SPAs in food tend to exist in the parent form. A significant difference between food categories was analyzed. The highest GM of ΣSPAs occurred in cereals and cereal products, which was approx. 8 times higher than that in beverages. The estimated daily intakes of ΣSPAs in Chinese preschoolers, school-age children, adults and elderly individuals were 22200, 9970, 7540 and 7700 ng/kg bw/day, resp. The exposure risks of SPAs decreased with age. This is the first simultaneous anal. of SPAs in multiple provinces and multiple categories of foodstuffs.

Bacterial resistance to antibiotics is an increasing threat to global healthcare systems. We therefore sought compounds with potential to reverse antibiotic resistance in a clin. relevant multi-drug resistant isolate of Escherichia coli (NCTC 13400). 200 natural compounds with a history of either safe oral use in man, or as a component of a traditional herb or medicine, were screened. Four compounds; ellagic acid, Pr gallate, cinchonidine and cepharanthine, lowered the min. inhibitory concentrations (MICs) of tetracycline, chloramphenicol and tobramycin by up to fourfold, and when combined up to eightfold. These compounds had no impact on the MICs of ampicillin, erythromycin or trimethoprim. Mechanistic studies revealed that while cepharanthine potently suppressed efflux of the marker Nile red from bacterial cells, the other hit compounds slowed cellular accumulation of this marker, and/or slowed bacterial growth in the absence of antibiotic. Although cepharanthine showed some toxicity in a cultured HEK-293 mammalian cell-line model, the other hit compounds exhibited no toxicity at concentrations where they are active against E. coli NCTC 13400. The results suggest that phytochems. with capacity to reverse antibiotic resistance may be more common in traditional medicines than previously appreciated, and may offer useful scaffolds for the development of antibiotic-sensitizing drugs.

We previously reported that lysine-demethylase 2A (KDM2A), a Jumonji-C histone demethylase, is activated by gallic acid to reduce H3K36me2 levels in the rRNA gene promoter and consequently inhibit rRNA transcription and cell proliferation in the breast cancer cell line MCF-7. Gallic acid activates AMP-activated protein kinase (AMPK) and increases reactive oxygen species (ROS) production to activate KDM2A. Esters of gallic acid, Pr gallate (PG) and epigallocatechin gallate (EGCG), and other chems., reduce cancer cell proliferation. However, whether these compounds activate KDM2A has yet to be tested. In this study, we found that PG and EGCG decreased rRNA transcription and cell proliferation through KDM2A in MCF-7 cells. The activation of both AMPK and ROS production by PG or EGCG was required to activate KDM2A. Of note, while the elevation of ROS production by PG or EGCG was limited in time, it was sufficient to activate KDM2A. Importantly, the inhibition of rRNA transcription and cell proliferation by gallic acid, PG, or EGCG was specifically observed in MCF-7 cells, whereas it was not observed in non-tumorigenic MCF10A cells. Altogether, these results suggest that the derivatization of gallic acid may be used to obtain new compounds with anti-cancer activity.

In this study, an optical thermometer based on regenerated cellulose fibers modified with YF3: 20% Yb3+, 2% Er3+ nanoparticles was developed. The presented sensor was fabricated by introducing YF3 nanoparticles into cellulose fibers during their formation by the so-called Lyocell process using N-methylmorpholine N-oxide as a direct solvent of cellulose. Under near-IR excitation, the applied nanoparticles exhibited thermosensitive upconversion emission, which originated from the thermally coupled levels of Er3+ ions. The combination of cellulose fibers with upconversion nanoparticles resulted in a flexible thermometer that is resistant to environmental and electromagnetic interferences and allows precise and repeatable temperature measurements in the range of 298-362 K. The obtained fibers were used to produce a fabric that was successfully applied to determine human skin temperature, demonstrating its application potential in the field of wearable health monitoring devices and providing a promising alternative to thermometers based on conductive materials that are sensitive to electromagnetic fields.

Extensive and long-term applications have resulted in global distributions of plastic additives (PAs). To facilitate the understanding of human exposure to PAs, the present study investigated the abundances and profiles of a broad range of PAs on human hands via hand wipe sampling. Sixty out of 160 PAs were detected in >50% of hand wipes collected from 30 children and 45 adults, among which a number of organophosphate esters (OPEs) and synthetic antioxidants that have rarely or never been investigated in prior studies. The total masses of PAs ranged from 650 to 87 030 ng (median: 6110 ng) and 1230 to 19360 ng (median: 5600 ng) in adultsâ€?and children′s hand wipes, resp. By categories, phthalates (PAEs) represented the most abundant group of PAs, followed by non-PAE plasticizers, UV stabilizers, OPEs, antioxidants, and bisphenol analogs. Children exhibited greater PA levels per square centimeter of hand surface, indicating elevated exposure compared with adults. Strong correlations existed for many PAs between adults and children or within each subpopulation, indicating close connections between the two subpopulations in the exposure profiles. The great complexity of plastic additives on hands raises the need for future investigations on human exposure pathways and potential health risks from the “cocktail” effects.
