Chemical Synthesis of Fucosylated Chondroitin Sulfate Oligosaccharides

Fucosylated chondroitin sulfates (FuCSs) are a unique type of polysaccharides occurring in sea cucumber that show a variety of biological activities. In particular, well-defined FuCS oligosaccharides, consisting of a trisaccharide repeating unit of beta-D-GalNAc(4,6-diS)-(1 -> 4)-[alpha-L-Fuc(2,4-diS)-(1 -> 3)]-beta-D-GlcUA, display potent anticoagulant activity via selective inhibition of the intrinsic tenase, which could be developed into anticoagulant drugs without bleeding risk. Herein, we report an effective approach to the synthesis of FuCS oligosaccharides, as demonstrated by the successful elaboration of FuCS tri-, hexa-, and nonasaccharides. The syntheses employ an orthogonally protected trisaccharide as a pivotal building block that can be readily converted into the donor and acceptor for glycosidic coupling. In addition, the internal patterns of protecting groups, involving N-trichloroacetyl for N-acetyl group, benzylidene and benzyl groups for sulfonated hydroxyl groups, and benzoyl and methyl esters for free hydroxyl and carboxylic acid, respectively, ensure stereoselective formation of the glycosidic linkages and sequential transformation into the desired FuCS oligosaccharides.

Two-step biodiesel production using high free fatty acid containing pig fat

The aim of the study was to produce biodiesel from animal oil as non-edible feedstock. Three different concentrations of oils were used to produce the biodiesel. The main characteristic of these oils was high-free fatty acid content. The oils were transesterified with acid and basic catalysts, respectively. Biodiesel yield of 90 wt.% was obtained by acid transesterification (9 wt.% HCl, 6:1 methanol: oil molar ratio, 60 degrees C, 46 h). Pre-esterification conditions were studied using different oils and acid catalysts: 0.6 wt.% HCl, 6:1 methanol: oil molar ratio at 60 degrees C for 4 h, and this resulted in oil with an acid value of less than 0.8%. Under similar transesterification conditions, alkali transesterification of esterified fats resulted in the yield of 97.2 wt.% esters. Biodiesel properties were determined and most of the properties were within EN 14214 and ASTM D6751. Biodiesel from animal fats is less stable for oxidation and this is due to the absence of natural antioxidants as compared to the biodiesel of vegetable origin. For this reason, biodiesel produced from animal fats might not be adequate to use at 100% pure in vehicles during cold weather.

New beta-ketophosphonates for the synthesis of prostaglandin analogues. 2 Phosphonates with bicyclo[3.3.0]octene and bicyclo[3.3.0]octane scaffolds linked to the beta-keto group

beta-Ketophosphonates, with the keto group linked to a bicyclo[3.3.0]oct(a)ene fragment, were synthesized starting from two diacids. These diacids were first transformed into internal anhydrides and one into a diester. The anhydrides and the diester were reacted with the lithium salt of dimethyl methanephosphonate to give two carboxy beta-ketophosphonates, an ester beta-ketophosphonate and a bis beta-ketophosphonate in good yield. The ester beta-ketophosphonate, obtained by two routes was used in the E-HEW selective olefination of a prostaglandin aldehyde with an alpha-side chain to give the 15-keto prostaglandin analogue in good yield. The compounds were characterized by elemental analysis, IR and high resolution H-1- and C-13-NMR spectroscopies.

A Novel Therapeutic Approach for Colorectal Cancer Stem Cells: Blocking the PI3K/Akt Signaling Axis With Caffeic Acid

Cancer stem cells (CSCs) have been identified in a multiple of cancer types and resistant to traditional cancer therapies such as chemotherapeutic agents and radiotherapy, which may destroy bulk tumor cells but not all CSCs, contributing to reformation tumor masses and subsequent relapse. Moreover, it is very difficult to effectively identify and eliminate CSCs because they share some common phenotypic and functional characteristics of normal stem cells. Therefore, finding better therapeutic strategies to selectively target CSCs might be helpful to reduce subsequent malignancies. In the present study, we found that caffeic acid effectively suppresses self-renewal capacity, stem-like characteristics, and migratory capacity of CD44(+) and CD133(+) colorectal CSCs in vitro and in vivo. In addition, we also revealed that PI3K/Akt signaling may be linked to multiple colorectal CSC-associated characteristics, such as radio-resistance, stem-like property, and tumorigenic potential. To the best of our knowledge, this is the first study demonstrating that caffeic acid effectively targets colorectal CSC populations by inhibiting the growth and/or self-renewal capacity of colorectal CSCs through PI3K/Akt signaling in vitro and in vivo.

Highly Stretchable, Self-Healable, and Adhesive Polyurethane Elastomers Based on Boronic Ester Bonds

Integrating stretchability, self-healability, and surface adhesive properties in a single material is attractive for expanding the applications in demanding fields. Herein, we have studied the metathesis kinetics of catechol-derived boronic esters and incorporated them into polyurethane (PU) networks to prepare a type of PU elastomeric vitrimers with excellent stretchability, healability, and adhesive properties. Two series of dynamic PU elastomers that have different cross-linking densities and varied fractions of catechol moieties were simply prepared by a solution casting method from the linear PUs containing pendent catechol groups and 1,4-phenylenediboronic acid. These elastomers possess excellent stretchability and moderate mechanical properties that can be readily manipulated by varying the crosslinking density and the number of residual catechol groups. Moreover, the elastomers are healable and reprocessable under mild conditions and show moderate adhesive capability. The effects of the cross-linking density and residual catechol fraction on the thermal relaxation profiles and adhesive strength of the elastomers were explored in detail. Finally, we have demonstrated the potential of this kind of dynamic PU elastomers for applications as stretchable, healable, and self-adhesive conducting composite materials.

Spectral, thermoanalytical characterizations, properties, engine and emission performance of complementary biodiesel-diesel-pentanol/octanol blends

Waste cooking oil (WCO) is a mixture of animal fats and used vegetable oils used at high temperatures. It can be obtained at almost no cost. In this work, valorisation of WCO for biodiesel production (WCOME) besides assessing the quality of its binary and ternary blends with Euro diesel and higher alcohols of pentanol and octanol were examined. WCOME satisfied both EN 14214 and ASTM D6751 standards concerning cetane number (53.9), kinematic viscosity at 40 degrees C (3.55 mm(2)/s), density (899.9 kg/m(3)) at 20 degrees C and iodine value (106.59 g iodine/100 g). These results are attributed to the excellent degree of unsaturation (110.58). Introducing higher alcohols to biodiesel-diesel blends has further improved their densities and cold flow properties. Furthermore, spectral studies including FT-IR, UV-vis and NMR and thermoanalytical characterizations including TGA and DSC analyses were studied to assess the influence of pentanol and octanol addition into biodiesel-diesel blends. Finally brake thermal efficiency and BSFC and 3 regulated emissions of CO, THC and NOx emissions were evaluated. The current study proved the positive impact of higher alcohols addition to the blends. This study supports the ongoing research on biodiesel synthesis from waste cooking oil in Turkey.

Nature-inspired remodeling of (aza)indoles to meta-aminoaryl nicotinates for late-stage conjugation of vitamin B-3 to (hetero)arylamines

Despite the availability of numerous routes to substituted nicotinates based on the Bohlmann-Rahtz pyridine synthesis, the existing methods have several limitations, such as the inevitable ortho-substitutions and the inability to conjugate vitamin B-3 to other pharmaceutical agents. Inspired by the biosynthesis of nicotinic acid (a form of vitamin B-3) from tryptophan, we herein report the development of a strategy for the synthesis of meta-aminoaryl nicotinates from 3-formyl(aza)indoles. Our strategy is mechanistically different from the reported routes and involves the transformation of (aza)indole scaffolds into substituted meta-aminobiaryl scaffolds via Aldol-type addition and intramolecular cyclization followed by C-N bond cleavage and re-aromatization. Unlike previous synthetic routes, this biomimetic method utilizes propiolates as enamine precursors and thus allows access to ortho-unsubstituted nicotinates. In addition, the synthetic feasibility toward the halo-/boronic ester-substituted aminobiaryls clearly differentiates the present strategy from other cross-coupling strategies. Most importantly, our method enables the late-stage conjugation of bioactive (hetero)arylamines with nicotinates and nicotinamides and allows access to the previously unexplored chemical space for biomedical research. Vitamin B3 derivatives display a range of biological activities. Here, the authors report the synthesis of meta-aminoaryl nicotinates, derivatives of vitamin B3, and their late-stage conjugation with (hetero)arylamines, ultimately expanding the chemical space for biomedical research.

DEHP-induce damage in grass carp hepatocytes and the remedy of Eucalyptol

The wide application of plastic products led to the wide exposure of plasticizer in environment. As a new environmental pollutant, plasticizers’ toxicity researches were far from enough in fish. In order to further explore these mechanisms, we used Diethylhexyl phthalate (DEHP), a common plasticizer, treated the grass carp hepatocytes, and selected Eucalyptol (EUC) to study its antagonistic effect on DEHP. The results showed that after DEHP exposure, oxidative stress level and inflammation in grass carp hepatocytes were increased, and then mRNA and protein expression of apoptosis related markers were increased significantly, leading to hepatocytes apoptosis. Moreover, AO/EB staining and Hoethst staining also showed that the number of apoptotic cells increased after DEHP exposure. It should be noted that both EUC pretreatment and EUC simultaneous treatment could alleviate the oxidative stress, levels of inflammatory factors and apoptosis induced by DEHP. In comparison, the effect of EUC simultaneous treatment was better. Our results showed that DEHP induced apoptosis in grass carp hepatocytes through oxidative stress and inflammation, while EUC could alleviate apoptosis by reducing oxidative stress and inflammation caused by DEHP. The innovation of this study was to explore the interaction between DEHP and EUC for the first time. This study found that DEHP could cause apoptosis in grass carp hepatocytes through oxidative stress and inflammation; EUC had a good antagonistic effect on a series of damage in grass carp hepatocytes caused by DEHP, and EUC pretreatment and simultaneous treatment had a certain effect, among which, simultaneous treatment had a better effect. This study enriched the theoretical mechanism of DEHP toxicity in fish hepatocytes, and put forward the methods to solve the toxicity of DEHP.

Quality Markers for Astragali Radix and Its Products Based on Process Analysis

Due to the complex nature of traditional medicines, quality control methods need to cover two aspects: compliance of raw materials with quality standards and process control. Astragali radix (AR), the roots of Astragalus mongholicus Bunge, was selected in this study as an example of a widely used traditional medicine in various formulations. Astragaloside IV (AG IV) and calycosin 7-O-beta-D-glucoside (CG) are used as the markers for the quality control of AR and its products in the Chinese Pharmacopoeia. However, in the raw materials, malic acid esters of the CG and acetate esters of the astragaloside are easily decomposed into CG and AG IV during storage and processing of AR to make extracts for various preparations. The thermal stability of the isoflavonoids and astragalosides in decoction was studied. The level of CG and astragalosides (AG I/AG II/AG IV) was strongly affected by prolonged heat during processing, while calycosin was stable in the conditions. Also the major astragalosides in AR could fully converted into AG IV which eventually reaches a stable level under certain conditions. With calycosin and AG IV as marker components, practical, reproducible, and precise methods were established and applied to the quality analysis of AR from its raw materials to its intermediates and products. This study demonstrates that a full chemical profiles analysis of the whole manufacturing process (from raw materials-intermediates/extracts-final product) is important to identify quality markers (Q-markers) and even to establish proper analysis methods for traditional Chinese medicine products.

Conversion of Unprotected Aldose Sugars to Polyhydroxyalkyl and C-Glycosyl Furans via Zirconium Catalysis

An efficient, zirconium-catalyzed conversion of unprotected aldose sugars with acetylacetone to polyhydroxyalkyl furans or C-glycosylfurans is reported. The furan products are formed in up to 93% yield using 5-10 mol % ZrCl4. Pentoses are readily converted at room temperature, while hexoses and their oligosaccharides require mild heating (i.e., 50 degrees C). Efficient conversions of glycolaldehyde, glyceraldehyde, erythrose, a heptose, and glucosamine are also demonstrated. This approach outpaces each of the previous Lewis acid-catalyzed methods in at least one the following ways: (i) lower catalyst loadings; (ii) reduced reaction temperatures; (iii) shorter reaction times; (iv) equimolar substrate stoichiometry; (v) expanded sugar scope; (vi) higher selectivities; and (vii) the use of an Earth-abundant Zr catalyst.
