The reaction mixture was cooled to RT and the suspended solids were collected by filtration, washed with THF (2 x 100 mL) and then re-suspended in a solution of NH3 in MeOH (0.7 M) and stirred vigorously. After 30 min the solid was collected by filtration, washed with NH3 in MeOH (0.7 M) and then dried in vacuo to afford methyl 3-((4-((4-((te/f-butoxycarbonyl)amino)naphthalen-1-yl)oxy)pyrimidin-2- yl)amino)-5-methoxybenzoate, Intermediate H5(P), as a beige solid (34.0 g, 92% purity by HPLC, 56%). To a suspension of Intermediate H5(P) (10.2 g, 92% pure, 18.2 mmol) in DCM (50 mL) was added TFA (10 mL, 130 mmol) dropwise and the resulting black solution kept at RT for 21 hr. An additional aliquot of TFA (5.0 mL, 67 mmol) was added and after a further 3 hr the reaction mixture was evaporated in vacuo. The residue was co-evaporated with toluene (100 mL) then with a solution of NH3 in MeOH (0.7 M, 2 x 100 mL), triturated with MeOH (100 mL) then the resulting solid was collected by filtration, washed with MeOH (50 mL) and dried in vacuo to afford the title compound, Intermediate H5, as a beige solid (7.9 g, 99%); R* 2.15 min (Method 2 acidic, 92% pure); m/z 417 (M+H)+, (ES+).
